Click on <strong>Bio</strong>Frac <strong>and</strong> click the OK button. You will now see <strong>Bio</strong>Frac fraction collector in the Devices in<br />

Setup box. The F1 Rack (12–13 mm tubes) is automatically selected. Click OK to choose.<br />

In the Gradient Pump section of the setup screen enter your buffer names. In the Buffer A field, type in<br />

2x PBS, pH 7.2. In the Buffer B field, type in 2x PBS + 250 mM Imidazole, pH 8.0.<br />

You are now ready to program the separation steps for your method. To program your method, press<br />

the Protocol icon on the tool bar.<br />

3. Program the Method Protocol<br />

From the Options pull-down menu, ensure that Use Volume (ml) is selected so that the programming base<br />

is Volume (Figure 8.21).<br />

Programming the fraction collector (<strong>Bio</strong>Frac fraction collector)<br />

From the left side of the screen, press the fraction collection icon (Figure 8.22).<br />

In the pop-up window that appears, choose Collection Windows.<br />

Make sure the correct rack type is displayed (F1 12-13 mm tubes).<br />

Click on the tab Collection/Windows.<br />
