Relaxarea endotelio-dependentă (RED) a<br />
muşchiului neted vascular, esenţială în hemodinamică,<br />
se bazează pe semnalizare complexă. Scopul nostru este<br />
să analizăm utilizarea acestor semnale în RED indusă de<br />
agonişti, dependentă de debit şi propagată longitudinal,<br />
mecanismele variind în funcţie de specie, vârstă, sex,<br />
status hormonal, teritoriul/calibrul vascular, factorul<br />
vasodilatator şi tonusul vascular, stresul oxidativ şi<br />
diverse condiţii patologice. Trecem în revistă elementele<br />
de progres în înţelegerea funcţiei şi disfuncţiei<br />
endoteliale, recent discutate de cercetători din domeniu:<br />
hiperpolarizarea intercelulară şi vasodilataţia (Garland<br />
CJ); rolurile divergente ale tipurilor de canale de K+<br />
dependente de Ca2+ (De Wit C); canalele TRP în<br />
funcţia cardiovasculară (Nilius B), organizarea<br />
moleculară a canalelor de calciu operate de rezerve<br />
(Barritt GJ, Serban DN); eterogenitatea semnalelor de<br />
calciu induse de aceticolină în endoteliu (Beny JL);<br />
presiunea intraluminală, endotelinele, izoformele<br />
ciclooxigenazei şi funcţiile receptorilor vasculari pentru<br />
angiotensină (Koller A, Bild W); izoformele NOX,<br />
speciile reactive ale oxigenului şi biologia vasculară<br />
(Touyz RM); disfuncţia endotelială indusă de stresul<br />
oxidativ (Forstermann U); profilul variabil al<br />
mecanismelor care asigură vasodilataţia endoteliodependentă<br />
în condiţii normale şi în diabet (Triggle CR,<br />
Serban IL); disfuncţia endotelială, rigiditatea vasculară<br />
şi calcificările ca modificări arteriale în boala renală<br />
cronică (Zoccali C, Covic AC). Raport sprijinit prin<br />

Abstract. Endothelium-dependent relaxation (EDR) of<br />
vascular smooth muscle, essential in hemodynamics, is<br />
based on complex signaling. Our goal is to analyze how<br />
such signals are used in EDR induced by vasoactive<br />
agents, and in flow-dependent and propagated<br />
vasodilation, while mechanisms vary with species, age,<br />
sex, hormonal status, vascular bed/caliber, the<br />
vasodilating factor and vascular tone, oxidative stress<br />
and pathological conditions. We review the elements of<br />
progress in understanding the mechanisms of endothelial<br />
functions and dysfunctions, as recently dicussed together<br />
by contributors in the field, as follows: intercellular<br />
hyperpolarization and vasodilation (Garland CJ);<br />
divergent roles for Ca-dependent K-channel types (De<br />
Wit C); TRP channels in cardiovascular function (Nilius<br />
B), molecular organization of store-operated calcium<br />
channels (Barritt GJ, Serban DN); heterogeneity of<br />
calcium responses to ACh in endothelial cells (Beny JL);<br />
intraluminal pressure, endothelins, COX isoforms, and<br />
functions of vascular angiotensin receptors (Koller A,<br />
Bild W); NOX homologues, reactive oxygen species,<br />
and vascular biology (Touyz RM); impairment of EDR<br />
by oxidative stress (Forstermann U); variable profile of<br />
mechanisms that ensure endothelium-dependent<br />
vasodilation in normal conditions and diabetes (Triggle<br />
CR, Serban IL); endothelial dysfunction, stiffness and<br />
calcifications as arterial changes in renal disease<br />
