Serum-free Systems<br />
PANEXIN NTA is a complete chemically defined serum<br />
substitute for the cultivation of adherent cells under serumfree<br />
conditions. PANEXIN NTA is developed with a unique<br />
technology and contains a special 3-dimensional substance<br />
release system (3D-SRS) for an optimal support of cells<br />
with nutrients and growth stimulants.<br />
The ready to use, sterile solution is added to the culture<br />
medium in a final concentration of 10%. It supports the<br />
adherent growth of many cell types in an optimum manner.<br />
Composition:<br />
PANEXIN NTA contains purified proteins, lipids, salts, amino<br />
acids, trace elements, attachment factors and hormones<br />
in an optimized formulation and a new 3-dimensional<br />
substance release system (3D-SRS). PANEXIN NTA contains<br />
no growth factors, undefined hydrolysates or peptones.<br />
Suitability:<br />
PANEXIN NTA is suitable for the cultivation of a variety of<br />
adherent cells under serum-free culture conditions.<br />
Special Advantages:<br />
It has been shown for many cell lines that PANEXIN NTA<br />
can fully replace FBS. Due to selected and pretested raw<br />
materials PANEXIN NTA batches are very homogeneous.<br />
Therefore the complex batch testing known from FBS can<br />
be omitted with the use of PANEXIN NTA. In addition, there<br />
is no need to change the previously used basal medium.<br />
PANEXIN NTA is completely chemically defined and contains<br />
no undefined peptones or hydrolysates. Therefore, the<br />
interpretation of results from studies on effects of<br />
individually added growth factors is easier and more reliable<br />
in serum-free conditions. For cell lines which require specific<br />
growth factors these should be added in a concentration<br />
as previously used.<br />
Instructions for Use :<br />
In many cases a serum-free cultivation can be done without<br />
complex a<strong>da</strong>ptation steps for adherent growing cell lines<br />
such as HEK293, CHO, L929, 3T3A.<br />
• Thaw PANEXIN NTA in a water bath at 37 °C. Please avoid<br />
repeated freeze-thaw cycles!<br />
• Trypsinate adherent cells as usual (e.g. 0.25% trypsin<br />
solution). Once the cells have become round and detach<br />
from the surface (the process can be speeded at 37 °C)<br />
inactivate trypsin with trypsin inhibitor.<br />
• Rinse the cells with DPBS (without Mg ++ /Ca ++ ) and centrifuge,<br />
sterile remove supernatant.<br />
• Resuspend cells in basal medium (e.g. RPMI 1640,<br />
DMEM or other) and count.<br />
• Add 10% sterile PANEXIN NTA to basal medium to<br />
replace FBS.<br />
• Add the cell suspension to the basal medium supplemented<br />
with PANEXIN NTA.<br />
• Initial seeding density 5,000 – 20,000 cells/cm 2 .<br />
• Incubate the cells in the usual way in a CO2 incubator<br />
at 37 °C.<br />
Depending on the cell type, the optimal PANEXIN NTA<br />
concentration can vary from 5% to 15%, comparable to FBS<br />
concentrations used. The optimal PANEXIN NTA concentration<br />
should be determined for each cell line. The tests should be<br />
started at a PANEXIN NTA concentration of 10% as in most<br />
cells the best results were obtained with this concentration.<br />

PANEXIN NTA <strong>ist</strong> ein komplett chemisch definierter<br />
Serumersatz zur Kultivierung von adhärenten Zellen unter<br />
serum-freien Bedingungen. PANEXIN NTA wurde mit einer<br />
besonderen Technologie entwickelt und enthält ein<br />
neuartiges 3-dimensio-nales Wirkstoff-Freisetzungssystem<br />
(3D-SRS), um Zellen optimal mit Nährstoffen zu versorgen.<br />
Die gebrauchsfertige, sterile Lösung wird in einer Endkonzentration<br />
von 10% dem Kulturmedium zugesetzt. Es unterstützt<br />
in optimaler Weise <strong>da</strong>s Wachstum vieler adhärenter Zelltypen.<br />
Zusammensetzung:<br />
PANEXIN NTA enthält aufgereinigte Proteine, Lipide, Salze,<br />
Aminosäuren, Spurenelemente, Anheftungsfaktoren und<br />
Hormone in einer optimierten Rezeptur. Es enthält keine<br />
Wachstumsfaktoren, undefinierte Hydrolysate oder Peptone.<br />
Eignung und Anwendungsgebiete:<br />
PANEXIN NTA <strong>ist</strong> geeignet für die Kultivierung einer Vielzahl<br />
von adhärenten Zellen unter serum-freien Bedingungen.<br />
Besondere Vorteile:<br />
PANEXIN NTA kann für viele Zelllinien anstelle von FBS<br />
verwendet werden. Durch ausgewählte und vorgetestete<br />
Rohstoffe sind die einzelnen Chargen sehr homogen und<br />
aufwändige Chargentestungen - wie sie bei FBS üblich<br />
sind - können entfallen. Das gewohnte Basismedium kann<br />
weiterhin verwendet werden. PANEXIN NTA <strong>ist</strong> komplett<br />
chemisch definiert und enthält keine undefinierten<br />
Hydrolysate oder Peptone. PANEXIN NTA enthält keine<br />
weiteren Zusätze an Wachstumsfaktoren. Untersuchungen<br />
zur Wirksamkeit von zugesetzten Wachstumsfaktoren<br />
werden <strong>da</strong>durch eindeutiger in ihrer Aussage. Für Zelllinien,<br />
die spezifische Wachstumsfaktoren benötigen, sollten<br />
diese weiterhin in der bisher verwendeten Konzentration<br />
zugegeben werden.<br />
Anwendung:<br />
In vielen Fällen kann eine serum-freie Kultivierung ohne aufwändige<br />
A<strong>da</strong>ptationsschritte durchgeführt werden (adhärent<br />
wachsende Zell-Linien wie z.B. HEK293, CHO, L929, 3T3A).<br />
• PANEXIN NTA im Wasserbad (37 °C) auftauen. (Bitte<br />
wiederholtes Auftauen vermeiden!)<br />
• Adhärente Zellen in gewohnter Weise abtrypsinieren<br />
(z.B. 0,25% Trypsinlösung). Sobald sich die Zellen abgerundet<br />
und von der Oberfläche abgelöst haben (der<br />
Vorgang kann bei 37 °C beschleunigt werden) mit<br />
Trypsininhibitor inaktivieren.<br />
• Zellen in PBS (ohne Mg ++ /Ca ++ ) aufnehmen und zentrifugieren.<br />
Überstand steril abnehmen und verwerfen.<br />
Zellen in Basismedium (RPMI 1640, DMEM oder<br />
andere) resuspendieren und die Zellzahl bestimmen.<br />
• Zum gewohnten Basismedium (z.B. RPMI 1640, DMEM,<br />
MEM, DMEM/F12) 10% PANEXIN NTA steril zugeben.<br />
• Zellsuspension zu dem mit PANEXIN NTA supplementierten<br />
Basismedium geben.<br />
• Einsaatdichte 5.000 – 20.000 Zellen/cm 2 .<br />
• Zellen in der gewohnten Weise im CO2-Brutschrank bei<br />
37 °C inkubieren.<br />
Je nach Zelltyp kann im Einzelfall die optimale PANEXIN<br />
NTA-Konzentration von 5% bis 15% variieren. Die optimale<br />
Konzentration für PANEXIN NTA sollte für jeden Zelltyp<br />
ermittelt werden. Bei 10% PANEXIN NTA-Konzentration<br />
können die Versuche gestartet werden, <strong>da</strong> mit dieser<br />
Konzentration bei den me<strong>ist</strong>en Zellen die besten Resultate<br />
erzielt wurden.<br />
