Serum-free Systems<br />
Panexin BMM is a chemically defined serum substitute<br />
for the cultivation of macrophages from mouse bone<br />
marrow (murine bone marrow derived macrophages, BMM)<br />
under serum-free conditions. The ready-to-use sterile solution<br />
in a final concentration of 5 % is added to the basalmedium<br />
RPMI 1640, supplemented with 50 μM Mercaptoethanol<br />
and 2 ng/ml GM-CSF mur. rec.<br />
Composition:<br />
Panexin BMM contains purified proteins, lipids, salts, amino<br />
acids, trace elements, attachment factors and hormones<br />
in an optimized formulation. It contains no growth factors,<br />
undefined hydrolysates or lysates (e. g. Peptones).<br />
Suitability:<br />
Panexin BMM has been developed for the generation of<br />
murine macrophages from bone marrow under serum-free<br />
conditions. This achieves stan<strong>da</strong>rdized conditions and reproducible<br />
results.<br />
Particular benefits:<br />
Panexin BMM allows the generation of murine macrophages<br />
from bone marrow under stan<strong>da</strong>rdized serum-free conditions.<br />
The results will be more comparable, as undefined<br />
components – like in serum cultures – are eliminated. In<br />
Panexin BMM matured macrophages will show excellent<br />
attachment capabilities.<br />
Application:<br />
For the production of complete serum-free medium, add<br />
5 % Panexin BMM to RPMI 1640, supplemented with<br />
50 μM Mercaptoethanol and 2ng/ml GM-CSF (murine, rec.).<br />
After the isolation of the bone marrow from a mouse, a<br />
single cell suspension will be achieved through frequent<br />
pipetting. Then centrifuge the cells at 200 x g for 10 minutes.<br />
Resuspend the isolated cells in the serum-free medium<br />
and seed the cells in three 75 mm 2 culture bottles within<br />
20 ml of medium. The incubation is done at 37° C and<br />
5 % CO2 fumigation in the incubator. The feeding of the<br />
cells should be done on <strong>da</strong>y 5 and 7 by an exchange of<br />
10 ml of medium with fresh medium. After 10 <strong>da</strong>ys, the<br />
cells can be harvested.<br />

Panexin BMM <strong>ist</strong> ein chemisch definierter Serumersatz<br />
zur Kultivierung von Makrophagen aus murinem Knochenmark<br />
(murine bone marrow derived macrophages, BMM)<br />
unter serumfreien Bedingungen. Die gebrauchsfertige<br />
sterile Lösung wird in einer Endkonzentration von 5 % zum<br />
Basalmedium RPMI 1640 zugegeben, ergänzt mit 50 μM<br />
Mercaptoethanol und 2 ng/ml GM-CSF mur. rec.<br />
Zusammensetzung:<br />
Panexin BMM enthält aufgereinigte Proteine, Lipide, Salze,<br />
Aminosäuren, Spurenelemente, Anheftungsfaktoren und<br />
Hormone in einer optimierten Rezeptur. Es enthält keine<br />
Wachstumsfaktoren und undefinierten Hydrolysate oder<br />
Lysate (z. B. Peptone).<br />
Eignung und Anwendungsgebiete:<br />
Panexin BMM <strong>ist</strong> für die Generierung von murinen Makrophagen<br />
aus Knochenmark unter serumfreien Bedingungen<br />
entwickelt worden. Dies erlaubt stan<strong>da</strong>rdisierte Bedingungen<br />
und reproduzierbare Ergebnisse.<br />
Besondere Vorteile:<br />
Panexin BMM ermöglicht die Generierung von murinen<br />
Makrophagen aus Knochenmark unter stan<strong>da</strong>rdisierten<br />
serumfreien Bedingungen. Die Ergebnisse werden <strong>da</strong>durch<br />
vergleichbar, <strong>da</strong> nicht definierte Komponenten wie sie bei<br />
serumhaltiger Kultur vorkommen, eliminiert werden. In<br />
Panexin BMM gereifte Makrophagen weisen eine exzellente<br />
Adhärenz auf.<br />
Anwendung:<br />
Zur Herstellung des kompletten serumfreien Mediums<br />
5 % Panexin BMM in RPMI 1640 geben, mit 50 μM Mercaptoethanol<br />
und 2ng/ml GM-CSF (murine, rec.) supplementieren.<br />
Nach der Isolierung des Knochenmarks aus einer Maus<br />
erhält man durch häufiges Auf- und Abpipettieren eine<br />
Einzelzellsuspension. Die Zellen werden anschließend bei<br />
200 x g 10 Minuten abzentrifugiert. Die isolierten Zellen<br />
eines Tieres werden im serumfreien Medium resuspendiert<br />
und in drei 75 mm 2 Kulturflaschen mit 20 ml Medium eingesät.<br />
Die Inkubation erfolgt bei 37° C und 5 % CO2-Begasung<br />
im Brutschrank. Eine Fütterung der Zellen erfolgt am<br />
5. und 7. Tag durch Austausch von jeweils 10 ml des Mediums<br />
mit frischem Medium. Nach 10 Tagen können die<br />
Zellen geerntet werden.<br />
