Serum-free Systems<br />
Preparation of PowerStem ESPro2 medium:<br />
PowerStem ESPro2 basal medium requires supplementation<br />
with PowerStem ESPro2 growth supplement and PowerStem<br />
ESPro2 LIF supplement. PowerStem ESPro2 complete medium<br />
(basal medium with supplement) is stable for 1 month when<br />
stored in the <strong>da</strong>rk at +2° C to +8° C.<br />
Instructions for use:<br />
• Prepare gelatine-coated plates by covering them with a<br />
0.2% gelatine solution for at least 10 min in the incubator.<br />
• mES-cells should always be kept at a relatively high cell<br />
density to maintain their pluripotency.<br />
• The subculture is best carried out from a sub-confluent<br />
culture (70%-80% confluence).<br />
• Individual colonies should not touch each other.<br />
• Too dense growth promotes the differentiation of cells<br />
and thus may cause the loss of pluripotency.<br />
• Trypsinate mES-cells in the usual procedure (e.g. 0.25%<br />
trypsin solution).<br />
• Once the cells have become round and detach from the<br />
surface (the process can be speeded at 37° C), resuspend<br />
them in DPBS by thoroughly pipetting up and down several<br />
times and centrifuge for 5 min at 180x g at room<br />
temperature.<br />
• Evacuate supernatant sterile and remove it.<br />
• Resuspend cell pellet in DPBS again by pipetting up and<br />
down several times.<br />
• Good dissociation of the cells is important, the goal being<br />
to obtain single cells or very small aggregates of only 2-3<br />
cells. Larger cell clumps should not remain.<br />
• Due to the serum-free formulation of PowerStem ESPro2,<br />
there is no trypsin inactivating effect of FBS; use trypsininhibitor<br />
to stop trypsin activity.<br />
• Count cells and plate them on gelatine-coated culture<br />
dishes in supplemented PowerStem ESPro2<br />
• Incubate the cells in the usual way in an incubator at<br />
37 °C and 5% CO 2 .<br />
• Feed cells with fresh PowerStem ESPro2 every second<br />
<strong>da</strong>y.<br />
• The cells should be split at ratios between 1:4 and 1:8<br />
depending on the growth rates.<br />
Please note: For differentiation studies LIF supplement must<br />
be omitted.<br />

Zubereitung von PowerStem ESPro2:<br />
Dem PowerStem ESPro2 Basal Medium müssen PowerStem<br />
ESPro2 Growth Supplement und <strong>da</strong>s PowerStem ESPro2 LIF<br />
Supplement zugegeben werden. PowerStem ESPro2 Komplettmedium<br />
(Basalmedium mit Supplementen) <strong>ist</strong> bei richtiger<br />
Lagerung (+2 °C bis +8 °C) einen Monat haltbar.<br />
Gebrauchsanweisung:<br />
• Bereiten Sie mit Gelatine beschichtete Kulturgefäße vor,<br />
indem Sie diese mit einer 0,2% Gelatinelösung bedecken<br />
und für 10 Minuten in den Inkubator geben.<br />
• mES-Zellen sollten immer in einer relativ hohen Zelldichte<br />
gehalten werden, um die Pluripotenz zu erhalten.<br />
• Die Subkultur wird am Besten von einer sub-konfluenten<br />
Kultur durchgeführt (70%-80% Konfluenz)<br />
• Einzelne Kolonien sollten sich gegenseitig nicht berühren.<br />
• Zu dichtes Wachstum fördert die Differenzierung der<br />
Zellen, wodurch ein Verlust der Pluripotenz entstehen<br />
könnte.<br />
• Trypsinieren Sie mES-Zellen mit dem gewohnten Verfahren<br />
(z. B. 0,25% Trypsinlösung).<br />
• Sobald die Zellen rund sind und sich von der Fläche<br />
ablösen (der Prozess kann bei 37° C beschleunigt werden),<br />
setzen Sie diese in DPBS, indem Sie sie einige Male<br />
gründlich auf und ab pipettieren und sie für fünf Minuten<br />
bei 180x g bei Raumtemperatur zentrifugieren<br />
• Entfernen Sie den Überstand unter sterilen Bedingungen.<br />
• Nehmen Sie die Zellpellets noch einmal in DPBS auf,<br />
indem Sie einige Male auf und ab pipettieren.<br />
• Eine gute Auftrennung der Zellen <strong>ist</strong> wichtig, <strong>da</strong>s Ziel sind<br />
einzelne Zellen oder sehr kleine Aggregate von nur<br />
2-3 Zellen. Es sollten keine größeren Zellklumpen vorhanden<br />
sein.<br />
• Aufgrund der serum-freien Formulierung von PowerStem<br />
ESPro2 gibt es keinen Inaktivierungseffekt von FBS für<br />
Trypsin; verwenden Sie deshalb Trypsin-Inhibitor, um die<br />
Trypsinaktivität vollständig zu neutralisieren.<br />
• Zählen Sie die Zellen und geben Sie diese mit PowerStem<br />
ESPro2 in Gelatine-beschichtete Kulturgefäße.<br />
• Inkubieren Sie die Zellen auf gewohnte Art und Weise in<br />
einem Inkubator bei 37 °C und 5% CO 2 .<br />
• Füttern Sie die Zellen jeden zweiten Tag mit frischem<br />
PowerStem ESPro2.<br />
• Abhängig von der Wachstumsphase sollten die Zellen im<br />
Verhältnis von 1:4 oder 1:8 geteilt werden.<br />
Bitte beachten Sie: Für Differenzierungsstudien muss <strong>da</strong>s<br />
LIF Supplement weggelassen werden.<br />
