Serum-free Systems
Panserin 801 is a serumfree ready to use medium for the
cultivation of human keratinocytes.
Composition:
MCDB-153 is used as the basal medium which must be
supplemented with the supplied supplements just before
the use. These supplements are:
• Epidermal Growth Factor (EGF)
• Insulin
• Hydrocortisone
• Ethanolamine
• Phosphoethanolamine
• Pituitary Extract (BPE)
Suitability:
Panserin 801 has been developed for the serumfree
cultivation of human keratinocytes. Panserin 801 selectively
supports the growth of human keratinocytes and
concurrently prevents the overgrowth with fibroblasts.
Application:
• Thaw the supplied supplements.
• Add the basal medium under sterile conditions.
• Trypsinate keratinocytes, then wash the cells and stop
the remaining trypsin activity with a trypsin inhibitor
(10 mg/ml).
• Wash the cells again and determined cell amount.
• Adjust the cell density at 3.000 – 5.000 cell/ml in the
following subcultivation (initial seeding density 1 - 5 x
10 6 cells/cm 2 ). Incubate keratinocytes with supplemented
Panserin 801 in an incubator.
• Cell culture dishes could be precoated alternatively
with attachment factors (collagen, fibronectin or natural/
synthetic fragments of these factors).

Panserin 801 ist ein gebrauchsfertiges Medium für die
serumfreie Kultivierung von humanen Keratinozyten.
Zusammensetzung:
Als Basismedium wird MCDB-153 verwendet, das kurz vor
Gebrauch mit den mitgelieferten Supplementen ergänzt
werden muss. Dies sind:
• Epidermaler Wachstumsfaktor (EGF)
• Insulin
• Hydrocortison
• Ethanolamin
• Phosphoethanolamin
• Hypophysenextrakt (BPE)
Eignung und Anwendungsgebiete:
Panserin 801 ist für die serumfreie Kultivierung von
humanen Keratinozyten entwickelt worden. Panserin 801
unterstützt selektiv das Wachstum von humanen Keratinozyten
und verhindert gleichzeitig ein Überwuchern der
Kultur mit Fibroblasten.
Anwendung:
• Mitgelieferte Supplemente auftauen.
• Unter sterilen Bedingungen Supplemente zum Basalmedium
hinzufügen.
• Keratinozyten abtrypsinieren, mit PBS waschen und
restliches Trypsin mit Trypsin-Inhibitor (10 mg/ml) abstoppen.
• Nach einem erneuten Waschschritt Zellzahl bestimmen.
Bei Subkultivierung Zelldichte auf 3000 bis 5000
Zellen/ml einstellen. (Initialkultur 1 - 5 x 10 6 /Zellen/
cm 2 ). Keratinozyten mit supplementiertem Panserin
801 Zellen im Brutschrak inkubieren.
• Alternativ Kulturschalen vorher mit Anheftungsfaktoren
(Collagen, Fibronectin oder natürlichen bzw. synthetischen
Fragmenten dieser Faktoren) beschichten.
