Serum-free Systems<br />
Production and serumfree cultivation of dendritic cells<br />
from mononuclear cells of peripheral blood (PBMC).<br />
Instructions:<br />
Separation of Blood with Separating Medium<br />
(Pancoll 1,077 g/ml)<br />
The blood samples should be processed as soon as possible<br />
after production in order to achieve optimal results. A storage<br />
of the blood samples for 24 hours at ambient temperature<br />
causes among other things a reduced output of<br />
lymphocytes, a change of the surface markers and a reduced<br />
response on mitogen stimulation.<br />
1) Add Pancoll (3 ml) into a suitable, sterile centrifuge<br />
tube under sterile conditions.<br />
2) Carefully coat the separating medium with the diluted<br />
blood sample (4 ml). Important: Don‘t mix the blood<br />
sample with Pancoll!<br />
3) Centrifugation at 400 x g for 30 - 40 minutes at<br />
18 - 20° C.<br />
4) After the centrifugation, carefully take off the upper<br />
phase (containing serum and platelets) with a pipette<br />
without mixing the interphase with the lymphocytes.<br />
5) Transfer the lymphocyte band into a new centrifuge<br />
tube with a new pipette. Here it is important to take<br />
off the whole material of the interphase with as little<br />
volume as possible.<br />
6) Add at least 3 volumes of a physiological saline solution<br />
(6 ml) to the lymphocytes.<br />
7) Carefully suspend the lymphocytes with a pipette.<br />
8) Centrifugation at 60 - 100 x g for 10 minutes at<br />
18 - 20° C.<br />
9) Reject the supernatant. Repeat washing step (point<br />
6 - 9).<br />
Serumfree Cultivation of Dentritic Cells<br />
in Panserin 416:<br />
After the last washing step the mononuclear cells are<br />
transferred with a cell density of 1 x 10 7 cells/ml into Panserin<br />
416. In order to remove non-adherent cells, the culture<br />
dishes are put into the incubator for 2 hours. Then<br />
the supernatant is carefully taken off and replaced by new<br />
Panserin 416. GM-CSF (800 U/ml) and interleukin-4 (500<br />
U/ml) are added as growth factors. The culture dishes are<br />
incubated for another 6 <strong>da</strong>ys in the incubator and every<br />
<strong>da</strong>y half of the medium is replaced by new medium which<br />
is supplemented by GMCSF and IL-4.<br />

PANSERIN 416/PANSERIN 416<br />
Gewinnung und serumfreie Kultivierung dendritischer Zellen<br />
aus mononucleären Zellen aus periphärem Blut (PBMC).<br />
Gebrauchsanweisung:<br />
Auftrennung von Blut mit Trennmedium<br />
(Pancoll 1,077 g/ml)<br />
Die Blutproben sollten sobald wie möglich nach der Gewinnung<br />
verarbeitet werden um optimale Ergebnisse zu<br />
erzielen. Eine Lagerung der Blutproben für 24 Stunden<br />
bei Raumtemperatur bewirkt unter anderem eine verminderte<br />
Ausbeute an Lymphozyten, eine Veränderung der<br />
Oberflächenmarker und eine verminderte Antwort auf<br />
Mitogen-Stimulation.<br />
1) Unter sterilen Bedingungen Pancoll (3 ml) in ein geeignetes,<br />
steriles Zentrifugenröhrchen vorlegen.<br />
2) Vorsichtig mit der verdünnten Blutprobe (4ml) die<br />
Trennlösung überschichten. Wichtig: Blutprobe und<br />
Pancoll nicht vermischen!<br />
3) Zentrifugation bei 400 x g für 30 - 40 Minuten bei<br />
18 - 20° C.<br />
4) Nach der Zentrifugation obere Phase (enthält Serum<br />
und Platelets) vorsichtig mit einer Pipette abnehmen,<br />
ohne die Interphase mit den Lymphocyten zu vermischen.<br />
5) Mit einer neuen Pipette die Lymphozyten-Bande in ein<br />
neues Zentrifugenröhrchen überführen. Wichtig <strong>da</strong>bei<br />
<strong>ist</strong>, <strong>da</strong>s gesamte Material der Interphase mit möglichst<br />
wenig Volumen zu entnehmen.<br />
6) Mindestens 3 Volumina einer physiologischen Salzlösung<br />
(6 ml) zu den Lymphozyten geben.<br />
7) Mit einer Pipette die Lymphozyten vorsichtig suspendieren.<br />
8) Zentrifugation bei 60 - 100 x g für 10 Minuten bei<br />
18 - 20° C.<br />
9) Überstand verwerfen. Waschschritt wiederholen (Punkt<br />
6 - 9).<br />
Serumfreie Kultivierung von dendritischen Zellen<br />
in Panserin 416:<br />
Die mononucleären Zellen werden nach dem letzten Waschschritt<br />
mit einer Zelldichte von ca. 1 x 10 7 Zellen/ml in<br />
Panserin 416 überführt. Um nicht adhärente Zellen zu entfernen,<br />
werden die Kulturgefäße für 2 Stunden in den Brutschrank<br />
gestellt. <strong>Der</strong> Überstand wird <strong>da</strong>nn vorsichtig abgezogen<br />
und mit neuem Panserin 416 ersetzt. Als Wachstumsfaktoren<br />
werden GM-CSF (10ng/ml) und Interleukin-4<br />
(10 ng/ml) zugesetzt. Die Kulturgefäße werden für weitere<br />
6 Tage im Brutschrank bebrütet, wobei jeden Tag die Hälfte<br />
des Mediums mit neuem Medium, supplementiert mit<br />
GMCSF und IL-4, ersetzt wird.<br />
