Instructions for use:<br />
A<strong>da</strong>ption to a protein-free culture<br />
Most CHO-cells can be directly transferred from a serum<br />
containing adherent culture into the protein-free suspension<br />
culture. In most cases the stable suspension culture is<br />
developing within approx. 2 weeks.<br />
Direct a<strong>da</strong>ptation to Panserin C6000:<br />
• Use adherent cells in the log phase of a serum<br />
containing culture (for example high glucose DMEM<br />
with 10% FBS).<br />
• Remove serum containing medium with a pipette.<br />
• Wash cell layer with DPBS without Ca/Mg.<br />
• Cover cell layer with trypsin / EDTA (0.25%, 0.02%)<br />
(about 2 ml per T25 flask).<br />
• Remove trypsin after about 1 minute.<br />
• Incubate the cells until they show a round figure and<br />
detach from the surface (about 5 minutes).<br />
• Transfer cells into DPBS and count the cell number.<br />
• Seed 5 x 10 4 - 1 x 10 5 cells/ml in preheated Panserin<br />
C6000. Use flasks for suspension culture (e.g. greiner<br />
bio-one Code 690 190).<br />
• Incubation at 37 °C and 5% CO2 in an incubator.<br />
For a few <strong>da</strong>ys the cells will stay in a lag-phase where no<br />
significant cell proliferation will occur. After about one<br />
week, however, the cells will be accustomed to the proteinfree<br />
medium and will have proliferated to 1x10 6 cells/ml.<br />
Transfer the cells every 3 to 4 <strong>da</strong>ys into fresh medium.<br />
(Seeding density 3-5 x 10 4 cells/ml).<br />
Generation time of CHO-cells in Panserin C6000: 16.5 h.<br />

Gebrauchsanweisung:<br />
A<strong>da</strong>ption an die protein-freie Kultur<br />
CHO-Zellen können me<strong>ist</strong> direkt aus der serum-haltigen<br />
adhärenten Kultur in eine protein-freie Suspensions-Kultur<br />
überführt werden. Dabei <strong>da</strong>uert es ca. 2 Wochen bis sich<br />
eine stabile Suspensions-Kultur entwickelt.<br />
Direkte A<strong>da</strong>ption an Panserin C6000:<br />
• Zellen einer serum-haltigen adhärenten Kultur<br />
• (z.B. DMEM high glucose mit 10 % FBS) aus der logarithmischen<br />
Wachstumsphase verwenden.<br />
• Serum-haltiges Medium mit Pipette abnehmen.<br />
• Zellrasen mit DPBS ohne Ca/Mg, waschen.<br />
• Zellrasen mit Trypsin/EDTA (0,25%, 0,02%) überschichten<br />
(ca. 2 ml auf T25 Flasche).<br />
• Trypsin nach ca. 1 min abnehmen.<br />
• Inkubation bis sich die Zellen abrunden und vom Untergrund<br />
lösen. (nach ca. 5 min).<br />
• Zellen in DPBS aufnehmen und Zellzahl bestimmen.<br />
• 5 x 10 4 - 1 x 10 5 Zellen/ml in vorgewärmtes Panserin<br />
C6000 einsäen. Kulturflaschen für Suspensionskultur<br />
verwenden (z.B. Greiner Bio-One, Code 690 190).<br />
• Inkubation bei 37 °C und 5% CO2 im Brutschrank.<br />
In den ersten Tagen tritt eine lag-Phase ein, wobei kaum<br />
eine Zellvermehrung stattfindet. Nach ca. 7 Tagen jedoch<br />
haben sich die Zellen an <strong>da</strong>s protein-freie Medium gewöhnt<br />
und sich auf 1 x 10 6 Zellen/ml vermehrt.<br />
Zellen alle 3 bis 4 Tage in frisches Medium umsetzen.<br />
(Einsaat 3 – 5 x 10 4 Zellen/ml).<br />
Generationszeit CHO in Panserin C6000: 16,5 h.<br />
