Panserin 604SPF is a ready to use complete medium for
the cultivation of transfected and non-transfected CHO
cells (Chinese Hamster Ovary) in suspension culture.
Composition:
Panserin 604SPF is a completely defined medium for the
cultivation of CHO cells in suspension culture. It contains
no protein and also no human or animal ingredients. This
allows a simple purification of the final products and the
use in critical applications such as drug production.
Suitability:
Cultivation of transfected and non-transfected CHO cells
in suspension culture (bioreactor) for the production of
recombinant proteins.
Special Advantages:
Panserin 604SPF contains no undefined lysates or herbal
hydrolysates. Panserin 604SPF allows short adaptation
phases and very good growth rates. Through an optimal
and balanced composition of the medium an aggregate
formation of the cells, as observed in many serumfree
media, could be largely avoided. Particularly suitable for
the production of recombinant proteins combined with a
easier purification of the final products due to the low
protein content of the medium.
Instructions:
In many cases the switch from serum-containing medium
to Panserin 604SPF can be done without any special adaption
procedures. For those cells which do not tolerate an
immediate switch we recommend a primary culture with
Panserin 604SPF supplemented with serum and than a
stepwise reduction of serum towards a pure Panserin
604SPF cultivation. This stepwise adaption will also be
supported by higher cell seeds. Cells should be seeded in
a density of at least 5x 10 4 cells/ml. After a few passages
in serumfree culture at lower growth rates the cells reach
high growth rates. For the successful transfer into serumfree
cultivation the vitality of the cells is an important factor.
Thus the cells should be transferred in the logarithmic
growth phase. According to our experience a transfer within
the stationary growth phase will have lower prospects of
success. In adherent cells it should be assured that – if
trypsin is used for detachment – the enzyme is completely
washed out or is inactivated by trypsin-inhibitors in order
for the serum to have no neutralizing effect.

Panserin 604SPF ist ein gebrauchsfertiges Komplettmedium
zur Kultivierung von transfizierten und nicht-transfizierten
CHO-Zellen (Chinese Hamster Ovary) in Suspensionskultur.
Zusammensetzung:
Panserin 604SPF ist ein vollständig definiertes Medium
für die Kultivierung von CHO-Zellen in Suspensionskultur.
Es enthält keine Proteine (ausgenommen rec. Insulin in
geringer Konzentration) und keine anderen Komponenten
humanen oder tierischen Ursprungs. Dies ermöglicht eine
einfache Aufreinigung der Endprodukte und den Einsatz
in sensiblen Produktionsbereichen.
Eignung und Anwendungsgebiete:
Kultivierung von transfizierten und nicht-transfizierten CHO-
Zellen in Suspensionskultur (Bioreaktor) zur Gewinnung
von rekombinanten Proteinen.
Besondere Vorteile:
Panserin 604SPF enthält keine undefinierten Lysate oder
pflanzliche Hydrolysate. Es zeichnet sich durch kurze Adaptionsphasen
und sehr gute Wachstumsraten aus. Durch
eine optimale und ausgewogene Zusammensetzung des
Mediums konnte eine Aggregatbildung der Zellen, wie man
sie bei vielen serumfreien Medien beobachten kann, weitgehend
vermieden werden. Besonders geeignet für die
Produktion von rekombinanten Proteinen. Leichte Aufreinigung
der Endprodukte da geringer Proteingehalt.
Gebrauchsanweisung:
Ein Wechsel von serumhaltigem Medium auf Panserin 604SPF
ist meist ohne besondere Adaption möglich. Für Zelllinien, die
ein solches einfaches Umsetzen in Panserin nicht vertragen,
empfehlen wir, die Zellen in Panserin 604SPF mit Serumzusatz
zu kultivieren und den Seruman-teil allmählich zu reduzieren.
Unterstützt wird dieses Verfahren durch höhere Titer bei der
Aussaat. Zellen sollten in einer Dichte von mindestens 5 x
10 4 /Zellen/ml eingesetzt werden. Nach einigen Passa-gen in
serumfreier Kultur bei niedrigeren Wachstumsraten, erreichen
die Zellen hohe Wachstumsraten. Für eine erfolgreiche Überführung
der Zellen in eine serum-freie Kultur spielt der Vitalitätszustand
eine entscheidende Rolle. Daher müssen die
Zellen in der logarithmischen Wachstumsphase entnommen
werden. Aus unseren Erfahrungen gelingt die Anzucht aus der
stationären Phase mit deutlich geringeren Erfolgsaussichten.
Zu beachten ist, dass nach einer Ablösung der Zellen mit
Trypsin das Enzym durch vollständiges Auswaschen entfernt
wird bzw. durch einen Trypsin-Inhibitor gehemmt wird, da der
im Serum vorhandene Neutralisationseffekt wegfällt.
