Special Processing of Serum Lots/Spezialaufbereitung von Serumchargen<br />
Sterile filtration<br />
The sterile filtration is performed via a vali<strong>da</strong>ted process.<br />
The raw material passes a series of filters with decreasing<br />
pore sizes. The last filtration step is done with a 0,2 μm<br />
pore size sterile filter.<br />
IgG extraction<br />
With a chromatographic method (affinity chromatography)<br />
the antibodies in the serum are near-completely removed<br />
(< 5 μg/ml). The biological activity of the serum is not affected.<br />
Active charcoal filtration<br />
Serum is heated in a water bath with dextran and activated<br />
charcoal. The activated charcoal, together with the substances<br />
bound up in it, is then removed by centrifugation and<br />
filtration.<br />
Dilipidisation<br />
Lipids are removed from serum by affinity chromatography.<br />
High-grade dialysis<br />
Serum is dialysed whit a 10,000 <strong>da</strong>lton exclusion membrane<br />
against a physiological saline solution.<br />
Heat inactivation<br />
Serum is heated for 30 minutes at 56° C in a water bath,<br />
whereby it is shaken softly several times.<br />
Gamma irradiation<br />
Serum is exposed to irradiation with at least 25 kGy.<br />
Sterilfiltration<br />
Die Sterifiltration erfolgt in einem validierten Prozess. Das<br />
Rohserum passiert eine Reihe von Filtern mit immer kleiner<br />
werdender Porengröße. Als letzter Filtrationsschritt steht<br />
ein Sterilfilter mit einer Porengröße von 0,2 μm.<br />
IgG-Extraktion<br />
Durch ein chromatographisches Verfahren (Affinitätschromatographie)<br />
werden die im Serum enthaltenen Antikörper<br />
weitgehend entfernt (< 5 μg/ml). Die biologische Aktivität<br />
des Serums wird <strong>da</strong>bei nicht beeinträchtigt.<br />
Aktivkohle-Filtration<br />
Serum wird im Wasserbad mit Dextran und Aktivkohle erwärmt.<br />
Anschließend wird die Aktivkohle mit den <strong>da</strong>ran<br />
gebundenen Substanzen abzentrifugiert.<br />
Delipidisierung<br />
Lipide werden aus dem Serum durch Affinitätschromatographie<br />
abgetrennt.<br />
HighGrade-Dialyse<br />
Serum wird mit einer 10.000 Dalton Ausschlussmembran<br />
gegen einen Puffer dialysiert.<br />
Hitzeinaktivierung<br />
Serum wird in einem Wasserbad bei 56° C für 30 Minuten<br />
unter Schütteln erhitzt.<br />
Gamma-Bestrahlung<br />
Serum wird einer Gamma-Bestrahlung von mindestens 25<br />
kGy ausgesetzt.<br />
