Desarrollo <strong>de</strong>l diagnóstico <strong>de</strong> virus, viroi<strong>de</strong>s...<strong>de</strong> papa. En general, <strong>de</strong>bido a las ventajas que ofreceesta técnica inmunoenzimática, todas las institucionescientíficas la aplican con resultados que se hanrevertido en diversas tomas <strong>de</strong> medidas prácticas parala agricultura.Por otra parte, las técnicas moleculares se utilizan enlos programas <strong>de</strong> mejoramiento <strong>de</strong> hortalizas, en el programa<strong>de</strong> cítricos, en los manejos integrados <strong>de</strong> plagas,en la búsqueda y evolución <strong>de</strong> nuevas fuentes <strong>de</strong> resistencia.A la vez, el <strong>de</strong>sarrollo <strong>de</strong>l diagnóstico ha posibilitadoempren<strong>de</strong>r estudios epifitiológicos y elaborarmetodologías <strong>de</strong> trabajo útiles para elevar la capacitación<strong>de</strong> los técnicos <strong>de</strong>l sistema <strong>de</strong> sanidad vegetal cubano.Finalmente se <strong>de</strong>be expresar que, aunque aún nose han introducido, existen los conocimientos sobre laregeneración y transformación genética <strong>de</strong>l cultivo <strong>de</strong>la papa, con los cuales las instituciones <strong>de</strong>dicadas a laobtención <strong>de</strong> varieda<strong>de</strong>s <strong>de</strong> papa pue<strong>de</strong>n contar con ungermoplasma nuevo que posee un gen <strong>de</strong> resistencia alPLRV susceptible <strong>de</strong> utilizarse en los programas <strong>de</strong>mejoramiento, a<strong>de</strong>más <strong>de</strong> la existencia <strong>de</strong>l genomaclonado <strong>de</strong> un begomovirus americano, a partir <strong>de</strong>lcual se han generado sondas útiles para la <strong>de</strong>tección ei<strong>de</strong>ntificación <strong>de</strong> otros miembros <strong>de</strong> este género queafectan a cultivos <strong>de</strong> interés económico, como tabacoy frijol.REFERENCIASAvrameas, S.: «Coupling of Enzymes to Proteins with Glutaral<strong>de</strong>hido»,Immunochemistry 6:43-52, EE.UU., 1969.Clark, M. Posteriormente se <strong>de</strong>scribe la enfermedad<strong>de</strong>l virus <strong>de</strong>l mosaico <strong>de</strong>l tabaco (TMV) con el nombreMosaikkrankheit, y se <strong>de</strong>mostró que se transmitía <strong>de</strong>plantas enfermas a sanas por inoculación <strong>de</strong> los extractos[Meyer, 1886]. En este sentido Iwanowski, consi<strong>de</strong>radoel padre <strong>de</strong> la virología, comprobó en 1892 queestos extractos se mantenían infectivos aun <strong>de</strong>spués <strong>de</strong>pasar por filtros bacterianos, lo que <strong>de</strong>scartaba la posibilidad<strong>de</strong> que esta patología pudiera ser causada porbacterias. Posteriormente Smith y Boncquet, en 1915,comprobaron que otras anomalías observadas podíantransmitirse por insectos. En general, el período <strong>de</strong> 1900a 1935 se <strong>de</strong>dicó a la <strong>de</strong>scripción <strong>de</strong> virosis por los síntomasy las anormalida<strong>de</strong>s citológicas reveladas por elmicroscopio electrónico, <strong>de</strong> forma tal que en 1939 logróconfirmarse la estructura <strong>de</strong>l TMV. En 1951 Markham,al realizar estudios con la técnica <strong>de</strong> cristalografía <strong>de</strong>rayos X, concluyó que el RNA <strong>de</strong> los virus está <strong>de</strong>ntro<strong>de</strong> la ca<strong>de</strong>na <strong>de</strong> proteínas. En 1956 Crick y Watson <strong>de</strong>finieronque la cubierta proteica está basada en numerosassubunida<strong>de</strong>s idénticas a lo largo <strong>de</strong> las estructurashelicoidales o cúbicas <strong>de</strong> los virus, y Wittmann yWittmann-Liebold, en 1966, establecieron la secuencia<strong>de</strong> 158 aminoácidos <strong>de</strong> la proteína <strong>de</strong> TMV, y <strong>de</strong>terminaronla ocurrencia <strong>de</strong> razas y la inducción <strong>de</strong> mutantesartificiales. A partir <strong>de</strong> este momento otros hombres<strong>de</strong> ciencia han contribuido cada vez más al <strong>de</strong>sarrollo<strong>de</strong> esta rama <strong>de</strong> la fitopatología, hasta llegar a nuestrosdías, en los cuales se imponen con mayor fuerza lastécnicas <strong>de</strong> avanzada y la biotecnología.fitosanidad/17
