6th International Congress of DipterologyComparison of synaptic organization of the mushroom bodycalyx with the lateral horn neuropile in the brain ofDrosophila melanogaster.Yasuyama, K.Department of Biology, Kawasaki Medical School, Kurashiki, Okayama 701-0192, JapanProjection neurons (PNs) of the antennal lobe, a primary sensoryneuropile, serve to integrate sensory information of different modalitiesand to transfer information to other areas of the insect brain. Presumedcholinergic PNs in the brain of Drosophila melanogaster, immunoreactiveto choline acetyltransferase (ChAT-ir), convey olfactory informationbetween the antennal lobe and the conspicuous neuropiles, mushroombodies (MBs), and finally terminate in an area called lateral horn (LH).The MB is regarded as a high-order integrative center required forolfactory learning and memory. The LH is a prominent but poorlyinvestigated diffuse neuropiles that have so far remained blank area in theinsect brain. We investigated the texture and synaptic connections ofChAT-ir PNs in the LH and, comparatively, in the MB calyces byimmuno-electron microscopy. We especially looked for differences in thepatterns of cholinergic fiber innervation between these two neuropiles, andfor general difference in the design of their neural circuits.The ChAT-ir PN terminals establish an array of large boutons (diameters2-7µm) throughout MB calyces. The ChAT-ir boutons form the core ateach glomerulus, and are encircled by tiny dendrites of the local intrinsicinterneurons, as well as by a number of GABAergic terminals. Thecholinergic calycal boutons are the largest identified cholinergic synapsesin the Drosophila brain. In the LH, the ChAT-ir PNs boutons accumulatein the lateral margins of the LH, where the more proximal LH exhibits lessintense immunolabeling. Synapses of ChAT-ir fibers appearpredominantly as divergent synaptic boutons (diameters 1-3µm),connected to unlabeled postsynaptic profiles, or alternatively as a minorityof tiny postsynaptic spines (diameters 0.05-05µm) among unlabeledprofiles. The synaptic circuits of the LH are far more complex than the320
