CHARACTERIZATION OF POLYMORPHISM TNF-α -308 G/A IN PATIENTS WITH<br />

CHRONIC HEPATITIS C.<br />

Introduction: The elimination or persistent infection by Hepatitis C virus (HCV) is<br />

associated with viral <strong>and</strong> host factors, as well as polymorphisms cytokine gene. The<br />

TNF-α cytokine is present in the beginning <strong>of</strong> the immune response against the virus<br />

C. The TNF-α gene is located on human chromosome 6p21.3, where several<br />

polymorphisms were identified <strong>and</strong> considered to affect the production <strong>of</strong> cytokine.<br />

The aim <strong>of</strong> this study was to characterize the polymorphism -308 G/A TNFα in<br />

patients HCV + . Methods <strong>and</strong> results: The samples were collected from 22 patients<br />

HCV + untreated with interferon <strong>and</strong> ribavirina at the Fundação de Medicina Tropical<br />

<strong>of</strong> Amazonas. The DNA was extracted according to the Brazol protocol followed by<br />

PCR amplification <strong>of</strong> the DNA fragment, subsequently visualized on agarose gel<br />

stained with 2.5% DNA Knowing Safe. The data distribution <strong>of</strong> the -308 G/A TNFα<br />

demonstrated that 22 patients infected with HCV, 16 (72.2%) had GG genotype, <strong>of</strong><br />

which 02 (12.5%) were diagnosed with liver cirrhosis, while 06 (27.3 %) were GA<br />

genotype <strong>and</strong> they had no diagnosis for liver cirrhosis. Conclusions: The data<br />

suggest an association between liver cirrhosis patients <strong>and</strong> 308 G/GTNF-α genotype,<br />

because it was more prevalent in this population.<br />
