INFECTION IN A MURINE MODEL OF DIABETES MELLITU<br />

Introdution: The high hyperglycemia observed in diabetic patients results in the<br />

advanced glycation endproducts (AGEs) which are able to binding in receptors<br />

present in the endothelial <strong>and</strong> immune cells that result in the induction <strong>of</strong> a variety<br />

<strong>of</strong> cytokines. As a consequence, several disorders have been described in the<br />

tissue matrix network such as the development <strong>of</strong> renal glomerulosclerosis <strong>and</strong><br />

the difficulties in the wound healing process in skin tissue. Considering that: 1)<br />

hypoinsulinemia-hyperglicemia (HH) condition is associated with impaired<br />

immune response to fungal infections, 2) infection in the diabetic foot is one <strong>of</strong> the<br />

severe predisposing factors to amputation, 3) the mechanisms involved in skin<br />

tissue wound healing during infection in diabetic patients are not fully understood;<br />

in the present study, we evaluated the local inflammatory response <strong>and</strong> the<br />

distribution <strong>of</strong> collagen <strong>and</strong> reticulin fibers in the footpad from HH-mice during<br />

dermatophitic infection.<br />

Matherial <strong>and</strong> Results: Male Swiss HH-mice, induced by alloxan (60mg/kg),<br />

were submitted to subcutaneous inoculation <strong>of</strong> Trichophyton ntagrophytes into<br />

the footpad. Non-infected HH-mice, free-HH infected-mice <strong>and</strong> non-infected <strong>and</strong><br />

free-HH mice were used as control groups. The mice were evaluated on days 7<br />

<strong>and</strong> 14 after the fungal inoculation. The footpad were collected <strong>and</strong> submitted to<br />

histological analyses by routine procedures for embedding in paraffin<strong>and</strong>. The<br />

sections were stained with hematoxylin-eosin (HE), periodic acid-Schiff staining<br />

(PAS), Masson trichrome <strong>and</strong> reticulin silver stain. Our results showed that the<br />

introduction <strong>of</strong> the fungus in the footpad in free-HH mice resulted in an acute<br />

inflammatory response followed by a granulomatous reaction; the deposition <strong>of</strong><br />

the collagen fibers <strong>and</strong> the thickness <strong>of</strong> reticular fibers were more intense on day
