conjugate with optimal modification. Equilibrate and load the column with 10 mM phosphate<br />

buffer, pH 8.0. Wash the column with equilibrating buffer and elute with the same<br />

buffer containing 100 mM NaCl (first) and 250 mM NaCl (last). Measure the F/P ratios of<br />

each fraction, and select the appropriate fractions.<br />

6. Alexa fluorochromes are available only as a protein labeling kit from Molecular Probes,<br />

Inc. (Eugene, Oregon, USA; www.probes.com). The reactive dye has a succinimidyl ester<br />

moiety that reacts with primary amines of proteins. The conjugation steps are similar to<br />

those for fluorescein isothiocyanate.<br />
