2.3. Cross-Blot<br />

1. Borate-Tween (BT): 50 mM Na 2B 4O 7, pH 9.3, 0.1 % (v/v) Tween 20. Store at 4°C.<br />

2. SCN-borate-Tween (SBT): 1 M NaSCN, 50 mM Na 2B 4O 7, pH 9.3, 0.1 % (v/v) Tween 20.<br />

Store at 4°C.<br />

2.4. Immunostaining<br />

1. PBS: 8.1 mM Na 2HPO 4, 1.5 mM KH 2PO 4, 2.7 mM KCl, 140 mM NaCl. Store at 4°C.<br />

2. PBS-Tween: 0.1% (v/v) Tween 20 in PBS. Store at 4°C.<br />

3. PTS: 8.1 mM Na 2HPO 4, 1.5 mM KH 2PO 4, 2.7 mM KCl, 0.5 M NaCl, 0.1% (v/v) Tween<br />

20. Store at 4°C.<br />

4. 10 mg/mL Bovine serum albumin (BSA): Store aliquots of 10 mL at –20°C.<br />

5. NC paper.<br />

6. Horseradish peroxidase conjugated antibody with specificity for the immunoglobulin<br />

isotype of the test antiserum.<br />

7. Chloronaphthol, solid. Store at –20°C. Caution: irritant to skin, eyes, and respiratory organs.<br />

Wear gloves, protective glasses, and clothing, especially when handling the solid substance<br />

(buffy crystals). May be purchased as tablets, which can be handled with lower risk.<br />

8. 30% Hydrogen peroxide: Store at 4°C. Caution: strong oxidant. Corrosive. Avoid contact<br />

with eyes and skin.<br />

2.5. Miscellaneous<br />

1. Electrophoresis apparatus for SDS-PAGE: Mini-Protean (Bio-Rad, Richmond, CA) or<br />

equivalent.<br />

2. Gradient mixer for 2 × 5 mL.<br />

3. Semidry blotting apparatus.<br />

3. Methods<br />

Most conveniently, this protocol may be carried out according to the following time<br />

schedule:<br />

Day 1: Casting two SDS gels.<br />

Day 2: Casting the stacking gels.<br />

Running SDS-PAGE.<br />

Blotting of the gels onto NC.<br />

Incubating one blot and of reference strips from both blots with antiserum.<br />

Cross-blotting.<br />

Reequilibration overnight.<br />

Day 3: Incubation with second antibody.<br />

Detection of spots by enzyme reaction.<br />

This schedule proved convenient in our laboratory. Other schedules are possible, as<br />

time values for antibody incubations and washing steps given are minimum requirements<br />

from our experience and may be different with other materials.<br />

3.1. SDS-PAGE<br />

3.1.1. Casting Gradient Gels<br />

Volumes are given for the Mini-Protean gel system (Bio-Rad) for gels of 83 ×<br />

55 × 1.5 mm.
