Develop<strong>in</strong>g solution:<br />

Citric acid stock solution 2 L<br />

Amido black 2 g<br />

Kodak PhotoFlo 600<br />

Note: Add amido black to stirred solution of citric acid stock, stir 30 m<strong>in</strong>, and then<br />

add the PhotoFlo.<br />

Reagent<br />

c. Crystal violet<br />

Crystal violet 0.1 g<br />

Distilled water 100 mL<br />

Ammonia<br />

Notes: Dissolve 0.1 g crystal violet <strong>in</strong> 100 mL distilled water. Stir until the solid is<br />

completely dissolved. Adjust pH to 7 to 8 with ammonia.<br />

Procedure<br />

1. Bake the article to be exam<strong>in</strong>ed at 100°C for 30 m<strong>in</strong>.<br />

2. Immerse the article <strong>in</strong>to the solution for 3 m<strong>in</strong>.<br />

3. R<strong>in</strong>se the article with distilled water and allow the surfaces to dry.<br />

d. Coomassie blue<br />

Sta<strong>in</strong><strong>in</strong>g solution<br />

Coomassie brilliant blue R250 0.44 g<br />

Glacial acetic acid 40 mL<br />

Methanol 200 mL<br />

Distilled water 200 mL<br />

Desta<strong>in</strong><strong>in</strong>g solution<br />

Glacial acetic acid 40 mL<br />

Methanol 200 mL<br />

Distilled water 200 mL
