pSELECT-GFP-LC3 - GFP-LC3 Expression Plasmid<br />

Description<br />

pSELECT-GFP-LC3 is a mammalian expression vector conta<strong>in</strong><strong>in</strong>g the<br />

human LC3B gene fused at its 5’ end to the green fluorescent prote<strong>in</strong><br />

(GFP) gene. The same plasmid is available with the GFP gene alone as a<br />

control. This control plasmid is called pSELECT-NGFP-zeo. Both plasmids<br />

are selectable <strong>in</strong> bacteria and mammalian cells with Zeoc<strong>in</strong> .<br />

Application<br />

Expression of the GFP-LC3 fusion gene allows to visualize autophagosome<br />

formation <strong>in</strong> real time <strong>in</strong> live cells. Dur<strong>in</strong>g autophagosome formation, GFP-<br />

LC3 is processed and recruited to the autophagosome membrane, where<br />

it can be imaged as cytoplasmic puncta by fluorescence microscopy (see<br />

picture). The percentage of GFP-LC3 positive cells can be determ<strong>in</strong>ed and<br />

is <strong>in</strong>dicative of autophagosome formation.<br />

Contents and Storage<br />

pSELECT-GFP-LC3 and pSELECT-NGFP-zeo are provided as 20 μg of<br />

lyophilized DNA. The plasmids can be purchased <strong>in</strong>dividually or together.<br />

They are shipped at room temperature and should be stored at -20°C.<br />

Both plasmids are provided with 4 pouches of E. coli Fast-Media ® Zeo (2<br />

TB and 2 Agar). For more <strong>in</strong>formation about Fast-Media ® , see pages 44-45.<br />

Autophagy is regulated at different levels. Two connected signal<strong>in</strong>g pathways encompass<strong>in</strong>g class III phosphatidyl<strong>in</strong>ositol 3-k<strong>in</strong>ase (PI3K) and<br />

mammalian target of rapamyc<strong>in</strong> (mTOR) play a central role <strong>in</strong> this regulation. Autophagy requires PI3K autophagy but is negatively regulated<br />

by mTOR. Thus <strong>in</strong>hibitors of PI3K act as autophagy <strong>in</strong>hibitors while <strong>in</strong>hibitors of mTOR act as autophagy <strong>in</strong>ducers.<br />
