Background<br />
Interleuk<strong>in</strong>-33 (IL-33) is a member of the IL-1 family, a group of cytok<strong>in</strong>es<br />
that play important roles <strong>in</strong> host defense, immune regulation and<br />
<strong>in</strong>flammation. IL-33 mediates its biological effects through ST2 (also known<br />
as IL1RL1), a receptor expressed on Th2 and mast cells. IL-33 and ST2 form<br />
a complex with IL-1R accessory prote<strong>in</strong> (IL-1RAcP), a signal<strong>in</strong>g receptor<br />
subunit that is also a member of the IL-1R complex. IL-33 signal<strong>in</strong>g leads to<br />
the activation of NF-kB and MAP k<strong>in</strong>ases, and the production of Th2associated<br />
cytok<strong>in</strong>es.<br />
Description<br />
HEK-Blue IL-33/IL-1b cells are designed to detect bioactive IL-33 and<br />
IL-1b (and IL-1a) by monitor<strong>in</strong>g the activation of the NF-kB and AP-1<br />
pathways. They were generated by stable transfection of HEK-Blue <br />
IL-1b cells with the IL1RL1 gene. They express a SEAP reporter gene<br />
under the control of the IFN-b m<strong>in</strong>imal promoter fused to five NF-kB<br />
and five AP-1 b<strong>in</strong>d<strong>in</strong>g sites.<br />
Stimulation of HEK-Blue IL-33/IL-1b cells with human IL-33 or IL-1b,<br />
but not with IL-18 nor TNF-a, activates the NF-kB and AP-1 pathways<br />
trigger<strong>in</strong>g the production of SEAP (figure 2). Levels of SEAP <strong>in</strong> the<br />
supernatant can be easily determ<strong>in</strong>ed with QUANTI-Blue .<br />
Stimulation of HEK-Blue IL-33/IL-1b cells with recomb<strong>in</strong>ant human<br />
IL-1b can be blocked by us<strong>in</strong>g the neutraliz<strong>in</strong>g antibody anti-hIL-1b-IgA.<br />
• Detection range for human IL-33: 0.5 - 100 ng/ml<br />
• Detection range for human IL-1b: 0.5 - 100 ng/ml<br />
HEK-Blue IL-33/IL-1b cells are resistant to blasticid<strong>in</strong>, Zeoc<strong>in</strong> and<br />
hygromyc<strong>in</strong> B.<br />
Contents and Storage<br />
HEK-Blue IL-33/IL-1b cells are grown <strong>in</strong> DMEM medium, 2mM<br />
L-glutam<strong>in</strong>e, 10% FBS supplemented with 30 μg/ml blasticid<strong>in</strong>, 100 μg/ml<br />
Zeoc<strong>in</strong> and 200 μg/ml HygroGold (ultrapure Hygromyc<strong>in</strong>). Each vial<br />
conta<strong>in</strong>s 5-7 x 10 6<br />
cells and is supplied with 1 mg blasticid<strong>in</strong>, 10 mg Zeoc<strong>in</strong> ,<br />
10 mg HygroGold , 50 mg Normoc<strong>in</strong> and 1 pouch of QUANTI-Blue Cells are shipped on dry ice.<br />
