Biosafety Features<br />
The LENTI-Smart kit has been designed to elim<strong>in</strong>ate the risk of<br />
replication recomb<strong>in</strong>ation through:<br />
- the use of multiple plasmids<br />
- the absence of HIV accessory genes and non-cod<strong>in</strong>g sequences<br />
- the use of self-<strong>in</strong>activ<strong>in</strong>g lentiviral expression plasmids.<br />
To further <strong>in</strong>crease the biosafety of LENTI-Smart derived lentiviral vectors,<br />
InvivoGen provides the LENTI-Smart NIL kit for the production of non<strong>in</strong>tegrat<strong>in</strong>g<br />
lentiviral vectors reduc<strong>in</strong>g the risk of <strong>in</strong>sertional mutagenesis.<br />
Contents and Storage<br />
Two LENTI-Smart kits are available:<br />
• LENTI-Smart (INT) for the generation of <strong>in</strong>tegrat<strong>in</strong>g lentiviral vectors<br />
• LENTI-Smart Starter Kit for the generation of <strong>in</strong>tegrat<strong>in</strong>g and non<strong>in</strong>tegrat<strong>in</strong>g<br />
lentiviral vectors.<br />
LENTI-Smart (INT) is available <strong>in</strong> 2 sizes, either 5 vials or 10 vials. Each<br />
vial allows the transfection of HEK 293T cells with a lentiviral expression<br />
plasmid <strong>in</strong> a 10-cm culture plate or a 75 cm 2 flask.. The LENTI-Smart <br />
Starter Kit conta<strong>in</strong>s 5 vials of LENTI-Smart (INT) and 5 vials of LENTI-<br />
Smart NIL (see below).<br />
All LENTI-Smart kits are provided with a vial of the control lentiviral<br />
expression plasmid. The LENTI-Smart vials are provided lyophilized, the<br />
control plasmid is provided as a liquid. Products are shipped at room<br />
temperature and should be stored at -20°C.<br />

Integration of a transgene <strong>in</strong>to the host genome poses safety concerns due to the potential risk of <strong>in</strong>sertional mutagenesis. To overcome this<br />
problem, non-<strong>in</strong>tegrat<strong>in</strong>g lentiviral vectors have been designed that comb<strong>in</strong>e the advantageous features of lentiviral vectors with episomal<br />
replication alleviat<strong>in</strong>g the risk of <strong>in</strong>sertional mutagenesis. InvivoGen provides LENTI-Smart NIL, the first kit designed for the generation of<br />
non-<strong>in</strong>tegrat<strong>in</strong>g lentiviral vectors.<br />
Description<br />
LENTI-Smart NIL comb<strong>in</strong>es the pLV-iVSV-G plasmid (see page 23) and<br />
the pLV-HELP-NIL D64 plasmid, precomplexed to the transfection reagent,<br />
LyoVec . pLV-HELP-NIL D64 expresses a mutant <strong>in</strong>tegrase (D64V) result<strong>in</strong>g<br />
<strong>in</strong> the generation of lentiviral vectors that are <strong>in</strong>tegration defective 1,2 .<br />
Non-<strong>in</strong>tegrat<strong>in</strong>g lentiviral vectors allow transient transgene expression <strong>in</strong><br />
divid<strong>in</strong>g cells and long-term expression <strong>in</strong> non-divid<strong>in</strong>g cells, such as neurons.<br />
LENTI-Smart NIL is provided with the control lentiviral plasmid,<br />
pLV-Green, that expresses a GFP gene.<br />

Duration of GFP expression us<strong>in</strong>g <strong>in</strong>tegrat<strong>in</strong>g or non-<strong>in</strong>tegrat<strong>in</strong>g (NIL) lentiviral<br />
vectors. Percentage of GFP positive HCT116 cells was assessed by FACS analysis,<br />
follow<strong>in</strong>g transduction with LENTI-Smart (INT)- or LENTI-Smart NIL-derived<br />
lentiviral vectors.<br />
