Biosafety Features
The LENTI-Smart kit has been designed to eliminate the risk of
replication recombination through:
- the use of multiple plasmids
- the absence of HIV accessory genes and non-coding sequences
- the use of self-inactiving lentiviral expression plasmids.
To further increase the biosafety of LENTI-Smart derived lentiviral vectors,
InvivoGen provides the LENTI-Smart NIL kit for the production of nonintegrating
lentiviral vectors reducing the risk of insertional mutagenesis.
Contents and Storage
Two LENTI-Smart kits are available:
• LENTI-Smart (INT) for the generation of integrating lentiviral vectors
• LENTI-Smart Starter Kit for the generation of integrating and nonintegrating
lentiviral vectors.
LENTI-Smart (INT) is available in 2 sizes, either 5 vials or 10 vials. Each
vial allows the transfection of HEK 293T cells with a lentiviral expression
plasmid in a 10-cm culture plate or a 75 cm 2 flask.. The LENTI-Smart
Starter Kit contains 5 vials of LENTI-Smart (INT) and 5 vials of LENTI-
Smart NIL (see below).
All LENTI-Smart kits are provided with a vial of the control lentiviral
expression plasmid. The LENTI-Smart vials are provided lyophilized, the
control plasmid is provided as a liquid. Products are shipped at room
temperature and should be stored at -20°C.

Integration of a transgene into the host genome poses safety concerns due to the potential risk of insertional mutagenesis. To overcome this
problem, non-integrating lentiviral vectors have been designed that combine the advantageous features of lentiviral vectors with episomal
replication alleviating the risk of insertional mutagenesis. InvivoGen provides LENTI-Smart NIL, the first kit designed for the generation of
non-integrating lentiviral vectors.
Description
LENTI-Smart NIL combines the pLV-iVSV-G plasmid (see page 23) and
the pLV-HELP-NIL D64 plasmid, precomplexed to the transfection reagent,
LyoVec . pLV-HELP-NIL D64 expresses a mutant integrase (D64V) resulting
in the generation of lentiviral vectors that are integration defective 1,2 .
Non-integrating lentiviral vectors allow transient transgene expression in
dividing cells and long-term expression in non-dividing cells, such as neurons.
LENTI-Smart NIL is provided with the control lentiviral plasmid,
pLV-Green, that expresses a GFP gene.

Duration of GFP expression using integrating or non-integrating (NIL) lentiviral
vectors. Percentage of GFP positive HCT116 cells was assessed by FACS analysis,
following transduction with LENTI-Smart (INT)- or LENTI-Smart NIL-derived
lentiviral vectors.
