pVITRO - Expression of Two Genes of Interest <strong>in</strong> vitro<br />

pVITRO is a family of plasmids developed ma<strong>in</strong>ly for <strong>in</strong> vitro studies. They allow the ubiquitous and constitutive co-expression of two genes of<br />

<strong>in</strong>terest. pVITRO plasmids can be stably transfected <strong>in</strong> mammalian cells and the genes of <strong>in</strong>terest are expressed at high levels. Each pVITRO<br />

plasmid is available with either two multiple clon<strong>in</strong>g sites or two reporter genes.<br />

Features and Benefits<br />

Gene Comb<strong>in</strong>ations <strong>in</strong> a S<strong>in</strong>gle Plasmid<br />

Co-expression of two or more genes from a s<strong>in</strong>gle vector is more efficient<br />

and convenient than us<strong>in</strong>g two separate vectors. pVITRO are multigenic<br />

plasmids that conta<strong>in</strong> two dist<strong>in</strong>ct transcription units (TU).<br />

Strong and Constitutive Expression of Two Transgenes<br />

Each pVITRO plasmid features two constitutive promoters that drive high<br />

levels of expression from two separate TU <strong>in</strong> a large number of<br />

mammalian cell l<strong>in</strong>es. Transcriptional <strong>in</strong>terference is m<strong>in</strong>imized by us<strong>in</strong>g<br />

promoters of different orig<strong>in</strong>s or promoters that are coord<strong>in</strong>ately<br />

activated (i.e. ferrit<strong>in</strong> promoters), and strong polyadenylation signals<br />

(polyA).<br />

• pVITRO1 plasmids carry two elongation factor 1 alpha (EF-1a)<br />

promoters, from rat and mouse orig<strong>in</strong>s comb<strong>in</strong>ed to the CMV and SV40<br />

enhancers respectively.<br />

• pVITRO2 plasmids conta<strong>in</strong> two human ferrit<strong>in</strong> composite promoters,<br />

FerH (heavy cha<strong>in</strong>) and FerL (light cha<strong>in</strong>), comb<strong>in</strong>ed to the SV40 and<br />

CMV enhancers respectively. To elim<strong>in</strong>ate the iron regulation, their 5’UTRs<br />

have been replaced by the 5’UTR of the mouse and chimpanzee EF-1a<br />

genes.<br />

S<strong>in</strong>gle Selection Marker for E. coli and Mammalian Cells<br />

pVITRO plasmids are available with different selectable markers that are<br />

active both <strong>in</strong> E. coli and mammalian cells: bsr (blasticid<strong>in</strong> resistance), hph<br />

(hygromyc<strong>in</strong> resistance), neo (kanamyc<strong>in</strong>/G418 resistance). In bacteria, the<br />

resistance gene is expressed from the E. coli EM7 promoter. In mammalian<br />

cells, it is transcribed from the promoter located 3’ of the Ori, as a<br />

polycistronic mRNA and translated through the IRES of the Foot and<br />

Mouth Disease Virus.<br />

Available with Two MCS or Two Reporter Genes<br />

pVITRO-mcs plasmids conta<strong>in</strong> two multiple clon<strong>in</strong>g sites (MCS) for the<br />

convenient clon<strong>in</strong>g of cDNAs.<br />

pVITRO-GFP/LacZ express the GFP and LacZ reporter genes, and can<br />

be used as controls. As the reporter genes are flanked by unique<br />

restriction sites, they can be used for the clon<strong>in</strong>g of open read<strong>in</strong>g frames<br />

that can be chosen from InvivoGen’s Gene A-List (see page 36 or visit<br />

our website www.<strong>in</strong>vivogen.com/orfs).<br />

Contents and Storage<br />

Each pVITRO plasmid is provided as 20 μg of lyophilized DNA. Product<br />

is shipped at room temperature and should be stored at -20°C. Plasmid<br />

is stable up to one year when properly stored.<br />

Each pVITRO is provided with 4 pouches of the appropriate E. coli Fast-<br />

Media ® (2 TB and 2 Agar).<br />
