Promoter CpG Methylation Study<br />

Methylation of CpG d<strong>in</strong>ucleotides with<strong>in</strong> the promoter region of genes is often associated with transcriptional<br />

silenc<strong>in</strong>g. This epigenetic event plays an important role <strong>in</strong> the regulation of gene activity <strong>in</strong> normal and cancer<br />

cells. InvivoGen provides pCpGfree-basic and pCpGfree-promoter, two SEAP reporter plasmids completely devoid<br />

of CpG d<strong>in</strong>ucleotides, that allow to study the effect of promoter CpG methylation <strong>in</strong> transfection assays. The lack<br />

of CpGs with<strong>in</strong> the plasmid backbone limits <strong>in</strong> vitro CpG methylation to the CpG d<strong>in</strong>ucleotides present <strong>in</strong> the<br />

<strong>in</strong>serted promoter fragment. Thus, the pCpGfree-basic and pCpGfree-promoter plasmids represent useful tools<br />

to analyze the effect of DNA methylation on CpG-conta<strong>in</strong><strong>in</strong>g promoters.<br />

Description<br />

The pCpGfree-basic and pCpGfree-promoter plasmids derive from the<br />

pCpGfree-mSEAP plasmid by deletion of the enhancer/promoter region<br />

or the enhancer sequence alone, respectively.<br />

pCpGfree-basic<br />

The pCpGfree-basic plasmid lacks the entire promoter region (i. e. mur<strong>in</strong>e<br />

CMV enhancer and human EF-1a promoter). It conta<strong>in</strong>s a multiple clon<strong>in</strong>g<br />

site upstream of the mur<strong>in</strong>e secreted embryonic alkal<strong>in</strong>e phosphatase<br />

(mSEAP) reporter gene. Expression of mSEAP <strong>in</strong> cells transfected with<br />

this plasmid depends on the <strong>in</strong>sertion of a functional promoter or<br />

enhancer/promoter cassette upstream from the mSEAP gene. Thus,<br />

pCpGfree-basic allows to study the effect of CpG methylation on a<br />

promoter, alone or comb<strong>in</strong>ed with enhancer elements.<br />

pCpGfree-promoter<br />

The pCpGfree-promoter plasmid conta<strong>in</strong>s the human EF-1a promoter<br />

and a multiple clon<strong>in</strong>g site <strong>in</strong> place of the mur<strong>in</strong>e CMV enhancer. It is<br />

specifically designed to analyze the effect of methylation on CpG residues<br />

present <strong>in</strong> enhancer elements.<br />

Procedure<br />

1- Insert promoter and/or enhancer fragment <strong>in</strong>to<br />

pCpGfree-basic or pCpGfree-promoter.<br />

2- Treat recomb<strong>in</strong>ant plasmid with CpG methylase (Sss I) or<br />

site specific-methylase (Hha I, Hpa II).<br />

3- Transiently transfect cells with methylase-treated or<br />

untreated plasmid.<br />

4- Analyze promoter CpG methylation by determ<strong>in</strong><strong>in</strong>g<br />

mSEAP expression us<strong>in</strong>g QUANTI-Blue , a SEAP detection<br />

medium.<br />

Contents and Storage<br />

pCpGfree-basic and pCpGfree-promoter plasmids are provided as 20 μg<br />

of lyophilized DNA with a paper disk of lyophilized E. coli GT115 stra<strong>in</strong>.<br />

Products are shipped at room temperature and should be stored at -20°C.<br />
