Blue Cytok<strong>in</strong>e Reporter Cells<br />

Blue Cytok<strong>in</strong>e Reporter Cells is an expand<strong>in</strong>g family of eng<strong>in</strong>eered cell l<strong>in</strong>es designed to provide a simple, rapid<br />

and reliable method to monitor the activation of signal<strong>in</strong>g pathways <strong>in</strong>duced by key cytok<strong>in</strong>es. They allow to detect<br />

these biologically active cytok<strong>in</strong>es and can also be used to screen for compounds exhibit<strong>in</strong>g agonist and antagonist<br />

activities. Blue Cytok<strong>in</strong>e Reporter Cells <strong>in</strong>clude B16-Blue and HEK-Blue Cells. B16-Blue and HEK-Blue Cells<br />

are eng<strong>in</strong>eered mur<strong>in</strong>e melanoma and HEK293 cells, respectively. They express an <strong>in</strong>ducible secreted embryonic<br />

alkal<strong>in</strong>e phosphatase (SEAP) reporter gene that can be quantitatively detected us<strong>in</strong>g QUANTI-Blue , a SEAP<br />

colorimetric detection medium. Blue Cytok<strong>in</strong>e Reporter Cells express this SEAP reporter gene under the control<br />

of specific promoters that are selectively activated by the cytok<strong>in</strong>es or other immune modulators known to <strong>in</strong>duce<br />

the pathway of <strong>in</strong>terest. Therefore, <strong>in</strong> the presence of the cytok<strong>in</strong>e, agonist or antagonist compound, the pathway<br />

is activated or <strong>in</strong>hibited modulat<strong>in</strong>g the SEAP activity. The amount of SEAP secreted <strong>in</strong> the cell supernatant can<br />

be measured spectrophotometrically with QUANTI-Blue .<br />
