pUNO-HA - TLR-HA Genes<br />

Features and Benefits<br />

pUNO-HA is a family of expression vectors featur<strong>in</strong>g HA-tagged TLR and IRF (Interferon<br />

Regulatory Factor) genes. The genes have been fused at the 3’end to the <strong>in</strong>fluenza<br />

hemaglut<strong>in</strong><strong>in</strong>e (HA) tag. This short sequence (YPYDVPDYA) encodes a peptide which is the<br />

epitope of a very efficient and specific monoclonal antibody. The use of HA-tagged TLR genes<br />

provides a simple and convenient method to detect the expression of the TLR genes by<br />

Western blot. All TLR genes can be detected us<strong>in</strong>g the same primary antibody, the HA tag<br />

monoclonal antibody (see page 73).<br />

pUNO-HA plasmids are selectable with blasticid<strong>in</strong> <strong>in</strong> both E. coli and mammalian cells.<br />

pUNO-TLR-GFP - TLR-GFP Fusion Genes<br />

TLR-GFP fusion prote<strong>in</strong>s can be used to study the localization of the TLRs. Transfected<br />

cells can be analyzed for GFP expression by flow cytometry and by Western-blott<strong>in</strong>g<br />

us<strong>in</strong>g GFP antibodies.<br />

Features and Benefits<br />

TLR-GFP fusion genes were generated by fus<strong>in</strong>g the GFP gene to the C term<strong>in</strong>us of various<br />

human TLR genes (TLR1 to TLR6). The TLR-GFP fusion genes are cloned <strong>in</strong> the pUNO<br />

plasmid under the control of the strong and ubiquitous mammalian promoter EF1a/HTLV.<br />

The pUNO plasmid is selectable with blasticid<strong>in</strong> <strong>in</strong> both E. coli and mammalian cells.<br />

All TLR::GFP fusion genes have been fully sequenced, their fluorescence confirmed and their<br />

function tested <strong>in</strong> HEK293 cells coexpress<strong>in</strong>g an NF-kB reporter plasmid and stimulated with<br />

the appropriate ligand.<br />

temperature and should be stored at -20ºC. Lyophilized E. coli cells are<br />

stable for at least 1 year when properly stored.<br />

Each plasmid is provided with 4 pouches of E. coli Fast-Media ® Blas (2 TB<br />

and 2 Agar, see pages 44-45).<br />
