pFUSE-Fc - Fc-Fusion Prote<strong>in</strong>s<br />

pFUSE-Fc is a family of plasmids developed to facilitate the construction of Fc-Fusion prote<strong>in</strong>s by fus<strong>in</strong>g a sequence encod<strong>in</strong>g a given prote<strong>in</strong><br />

to the Fc region of an immunoglobul<strong>in</strong>. The Fc-fusion prote<strong>in</strong>s are called immunoadhes<strong>in</strong>s when the Fc region is comb<strong>in</strong>ed with the functional<br />

doma<strong>in</strong> of a b<strong>in</strong>d<strong>in</strong>g prote<strong>in</strong>, such as a receptor, ligand or cell-adhesion molecule. The Fc region comprises the CH2 and CH3 doma<strong>in</strong>s of the<br />

IgG heavy cha<strong>in</strong> and the h<strong>in</strong>ge region. The h<strong>in</strong>ge serves as a flexible spacer between the two parts of the Fc-Fusion prote<strong>in</strong>, allow<strong>in</strong>g each part<br />

of the molecule to function <strong>in</strong>dependently.<br />

Description<br />

pFUSE-Fc<br />

pFUSE-Fc plasmids are designed for the generation of Fc-fusion prote<strong>in</strong>s. They feature<br />

a large choice of Fc regions from different species, with or without <strong>in</strong>trons, native or<br />

eng<strong>in</strong>eered. The gene encod<strong>in</strong>g the Fc region is cloned under the control of a strong<br />

and ubiquitous promoter. A multiple clon<strong>in</strong>g site (MCS) located upstream of the Fc<br />

region facilitates the clon<strong>in</strong>g of the functional doma<strong>in</strong> of a given prote<strong>in</strong>.<br />

pINFUSE-Fc<br />

pINFUSE-Fc is a new family of pFUSE-Fc plasmids featur<strong>in</strong>g genomic Fc regions. The<br />

presence of <strong>in</strong>trons is known to enhance the level of gene expression as splic<strong>in</strong>g is<br />

known to promote rapid and efficient mRNA export.<br />

pFUSE-SEAP-Fc<br />

pFUSE-SEAP-Fc plasmids are control plasmids that express the secreted embryonic<br />

alkal<strong>in</strong>e phosphatase (SEAP) reporter gene fused to each Fc region provided by<br />

InvivoGen.<br />

pFUSE-Fc, pINFUSE-Fc and pFUSE-SEAP-Fc plasmids are selectable with Zeoc<strong>in</strong> <strong>in</strong><br />

E. coli and mammalian cells. They can be used for transient or stable transfection.<br />

Features<br />

Secretion of Fc-Fusion Prote<strong>in</strong>s<br />

Two versions of pFUSE-Fc and pINFUSE-Fc exist to allow secretion of the Fc fusion:<br />

• No IL2ss: without the IL2 signal sequence (IL2ss) for the secretion of an Fc-fusion<br />

prote<strong>in</strong> conta<strong>in</strong><strong>in</strong>g the functional doma<strong>in</strong> of a prote<strong>in</strong> that is naturally secreted and<br />

thus conta<strong>in</strong>s a native signal sequence.<br />

• With IL2ss conta<strong>in</strong>s the IL2 signal sequence (IL2ss) for the generation of Fc-Fusion<br />

prote<strong>in</strong>s derived from prote<strong>in</strong>s that are not naturally secreted.<br />

Ease of Detection<br />

Fc-Fusion prote<strong>in</strong>s can be easily detected <strong>in</strong> the supernatant of pFUSE-Fc-transfected cells<br />

by SDS-PAGE. Their functional doma<strong>in</strong>s can be identified by immunoblott<strong>in</strong>g and ligand<br />

blott<strong>in</strong>g.<br />

Ease of Purification<br />

Fc-Fusion prote<strong>in</strong>s can be easily purified by s<strong>in</strong>gle-step prote<strong>in</strong> A or prote<strong>in</strong> G aff<strong>in</strong>ity<br />

chromatography.<br />

Contents and Storage<br />

All pFUSE-Fc, pINFUSE-Fc and pFUSE-SEAP-Fc plasmids are provided as 20 μg of lyophilized<br />

DNA. Product is shipped at room temperature and should be stored at -20°C. Plasmid is<br />

stable up to one year when properly stored. Each plasmid is provided with 4 pouches of<br />

E. coli Fast-Media ® Zeo (2 TB and 2 Agar, see pages 44-45).<br />
