Plasmoc<strong>in</strong> - The Mycoplasma Removal Agent<br />

Description<br />

Plasmoc<strong>in</strong> is a well-established antimycoplasma reagent. It conta<strong>in</strong>s two<br />

bactericidal components strongly active aga<strong>in</strong>st mycoplasmas that allow<br />

their elim<strong>in</strong>ation <strong>in</strong> only 2 weeks. The first component acts on the prote<strong>in</strong><br />

synthesis mach<strong>in</strong>ery while the second acts on the DNA replication. These<br />

two specific and separate targets are found only <strong>in</strong> mycoplasmas and<br />

many other bacteria and are completely absent <strong>in</strong> eukaryotic cells.<br />

In contrast to other anti-mycoplasma compounds, Plasmoc<strong>in</strong> is active<br />

on both free mycoplasmas and <strong>in</strong>tracellular forms. This advantage is<br />

conferred by one component of Plasmoc<strong>in</strong> which is actively transported<br />

<strong>in</strong>to mammalian cells. It ensures that follow<strong>in</strong>g treatment with Plasmoc<strong>in</strong> <br />

a cell culture is not re<strong>in</strong>fected by mycoplasmas released from <strong>in</strong>tracellular<br />

compartments of <strong>in</strong>fected cells.<br />

In all animal cell l<strong>in</strong>es tested to date, even at five times the work<strong>in</strong>g<br />

concentration, no apparent adverse effect on cellular metabolism is<br />

observed.<br />

No resistance <strong>in</strong> liquid cultures of mycoplasmas has ever been identified<br />

<strong>in</strong> repeated experiments attempt<strong>in</strong>g to measure the mutation rate.<br />

Therefore, development of resistant mycoplasma stra<strong>in</strong>s is virtually<br />

elim<strong>in</strong>ated.<br />

Plasmoc<strong>in</strong> is also active at low concentrations on a broad range of Gram<br />

positive and Gram negative bacteria that are otherwise resistant to the<br />

mixture of streptomyc<strong>in</strong> and penicill<strong>in</strong>, and exhibits no toxicity <strong>in</strong><br />

eukaryotic cells.<br />

Many cell l<strong>in</strong>es <strong>in</strong>fected by mycoplasmas have been successfully treated<br />

with Plasmoc<strong>in</strong> , <strong>in</strong>clud<strong>in</strong>g embryonic stem cells, hybridomas and<br />

retrovirus packag<strong>in</strong>g cells.<br />

• Plasmoc<strong>in</strong> Treatment<br />

To elim<strong>in</strong>ate mycoplasmas use Plasmoc<strong>in</strong> (ant-mpt) at<br />

25 μg/ml for two weeks <strong>in</strong> the <strong>in</strong>fected culture.<br />

• Plasmoc<strong>in</strong> Prophylactic<br />

To prevent mycoplasma contam<strong>in</strong>ation, use Plasmoc<strong>in</strong> <br />

(ant-mpp) at 2.5 - 5 μg/ml on a regular basis <strong>in</strong> cell culture.<br />

Plasmocure - The Alternative Mycoplasma Removal Agent<br />

Description<br />

In very rare cases, mycoplasmas resistant to Plasmoc<strong>in</strong> have been<br />

reported. To eradicate these mycoplasmas, InvivoGen has developed a<br />

new antimycoplasma agent called Plasmocure . Plasmocure comb<strong>in</strong>es<br />

two antibiotics that act through different mechanisms of action than those<br />

<strong>in</strong> Plasmoc<strong>in</strong> . A two week treatment with Plasmocure was found<br />

sufficient to completely elim<strong>in</strong>ate the mycoplasmas. A moderate toxicity<br />

can be observed dur<strong>in</strong>g the course of the treatment but full recovery of<br />

the cell l<strong>in</strong>e is expected once mycoplasmas are elim<strong>in</strong>ated.<br />

Plasmocure is a sterile ready-to-use solution. Simply add to mycoplasma<br />

contam<strong>in</strong>ated cell cultures for 2 weeks at the recommended<br />

concentration of 50 μg/ml.<br />

Contents and Storage<br />

Plasmoc<strong>in</strong> is provided as a yellow solution either at a concentration of 25<br />

mg/ml (Plasmoc<strong>in</strong> Treatment) or 2.5 mg/ml (Plasmoc<strong>in</strong> Prophylactic). One<br />

ml Plasmoc<strong>in</strong> Treatment is enough to treat 1 L medium. Plasmoc<strong>in</strong> is<br />

shipped at room temperature. Store at -20°C. Plasmoc<strong>in</strong> is stable 6 months<br />

at 4°C and at least one year at -20°C.<br />
