Gene A-List is a collection of high quality, full-length sequenced human and mouse open read<strong>in</strong>g frames (ORFs) available <strong>in</strong> an expression<br />
vector ready-to-use for prote<strong>in</strong> expression. All ORFs are cloned downstream of a strong and ubiquitous mammalian promoter which makes<br />
these clones suitable for expression and functional studies <strong>in</strong> various mammalian cell l<strong>in</strong>es.<br />

Full-length Sequenced Open Read<strong>in</strong>g Frames<br />
InvivoGen provides a large collection of human and mur<strong>in</strong>e genes, fully sequenced. These genes are provided as open read<strong>in</strong>g frames (ORFs) from the ATG<br />
to the Stop codon, exclud<strong>in</strong>g <strong>in</strong>trons and untranslated regions. As the ORFs are amplified from cDNA banks, a variant form is sometimes obta<strong>in</strong>ed. Most<br />
of the variations have been reported <strong>in</strong> Genbank. Furthermore for clon<strong>in</strong>g convenience, a neutral am<strong>in</strong>o acid is sometimes added immediately after the<br />
Start codon <strong>in</strong> order to create a suitable clon<strong>in</strong>g site.<br />
Among the Gene A-List , some genes encode prote<strong>in</strong>s that are naturally secreted. Their sequence <strong>in</strong>clude their native signal sequence which is generally<br />
located at the 5' end of the cod<strong>in</strong>g sequence. Other genes that code for eng<strong>in</strong>eered prote<strong>in</strong>s, such as particular fragments of longer prote<strong>in</strong>s (i.e. angiostatic<br />
prote<strong>in</strong>s) may <strong>in</strong>clude the signal sequence of the human IL-2 gene between the ATG and the second codon to allow their secretion.<br />
Suitable for Expression <strong>in</strong> Mammalian Cells<br />
Each ORF is cloned <strong>in</strong> a mammalian expression cassette consist<strong>in</strong>g of a potent and ubiquitous composite promoter and the strong SV40 polyadenylation<br />
signal. Each ORF is flanked by a unique restriction site at the 5’ end and 3’ end to facilitate its subclon<strong>in</strong>g <strong>in</strong>to another vector.<br />
The ORFs are supplied <strong>in</strong> two different plasmid backbones:<br />
• pORF, a plasmid selectable only <strong>in</strong> E. coli with ampicill<strong>in</strong><br />
• pBLAST/pUNO, a plasmid selectable <strong>in</strong> mammalian cells and <strong>in</strong> E. coli with blasticid<strong>in</strong>.<br />
