pNiFty - PRR Signal<strong>in</strong>g Reporter Plasmids<br />

PRR activation triggers a complex signal<strong>in</strong>g cascade that leads to the activation of different transcription factors, each play<strong>in</strong>g an important role <strong>in</strong> the<br />

subsequent immune response. To monitor the <strong>in</strong>duction of PRR signal<strong>in</strong>g <strong>in</strong> response to ligand stimulation <strong>in</strong> a simple and efficient manner, InvivoGen<br />

has designed pNiFty, a family of reporter plasmids express<strong>in</strong>g a reporter gene under the control of a m<strong>in</strong>imal promoter <strong>in</strong>ducible by these different<br />

transcription factors, either <strong>in</strong>dividually or <strong>in</strong> comb<strong>in</strong>ation. Most pNiFty plasmids are selectable with Zeoc<strong>in</strong> <strong>in</strong> both E. coli and mammalian cells, and<br />

can be used to generate stable clones<br />

pNiFty plasmids are composed of three key elements: a proximal<br />

promoter, repeated transcription factor b<strong>in</strong>d<strong>in</strong>g sites (TFBS) and a<br />

reporter gene. The proximal promoters are shorter than 500 bp and<br />

conta<strong>in</strong> transcription factor b<strong>in</strong>d<strong>in</strong>g sites. Upon stimulation <strong>in</strong> 293 cells,<br />

their expression level rema<strong>in</strong>s undetectable. With the addition of repeated<br />

TFBS, the proximal promoters become <strong>in</strong>ducible by the appropriate<br />

stimulus and drive the expression of the reporter gene.<br />

M<strong>in</strong>imal promoters<br />

- ELAM promoter: the proximal promoter of the endothelial cellleukocyte<br />

adhesion molecule (ELAM-1; E-select<strong>in</strong>) gene conta<strong>in</strong>s three<br />

NF-kB sites and is truly NF-kB specific, as it lacks an AP1/CREB site found<br />

<strong>in</strong> the full-length promoter 1, 2 .<br />

- IFN-b promoter: the mouse IFN-b m<strong>in</strong>imal promoter comprises several<br />

positive regulatory doma<strong>in</strong>s that b<strong>in</strong>d different cooperat<strong>in</strong>g transcription<br />

factors such as NF-kB, IRF3 and IRF7 3 .<br />

- ISG-56K promoter: the m<strong>in</strong>imal promoter of the human <strong>in</strong>terferonstimulated<br />

gene ISG-56K conta<strong>in</strong>s two <strong>in</strong>terferon-stimulated regulatory<br />

element (ISRE) sites and is fully <strong>in</strong>ducible by type I IFNs and <strong>in</strong>terferon<br />

regulatory factors (IRFs) 4, 5 .<br />

Transcription factor b<strong>in</strong>d<strong>in</strong>g sites (TFBS)<br />

- AP-1 b<strong>in</strong>d<strong>in</strong>g site: Activator prote<strong>in</strong> 1 (AP-1) is a transcription factor<br />

activated by most PRRs. AP-1 is a heterodimeric complex composed of<br />

members of Fos, Jun and, ATF prote<strong>in</strong> families. AP-1 b<strong>in</strong>ds to the TPA<br />

responsive element (TRE: ; TGAG/CTCA) 6 . AP-1 activation <strong>in</strong> TLR signal<strong>in</strong>g<br />

is mostly mediated by MAP k<strong>in</strong>ases such as c-Jun N-term<strong>in</strong>al k<strong>in</strong>ase (JNK),<br />

p38 and extracellular signal regulated k<strong>in</strong>ase (ERK).<br />
