TLR/NOD Ligand Screen<strong>in</strong>g<br />

InvivoGen has developed a high-throughput method to determ<strong>in</strong>e whether a compound is recognized by Toll-Like Receptors or NOD1/NOD2<br />

and acts either as an agonist or antagonist. This method is based on TLR- or NOD-<strong>in</strong>duced activation of various transcription factors <strong>in</strong>clud<strong>in</strong>g<br />

NF-kB, AP1 and IRF7 <strong>in</strong> HEK293 clones.<br />

Screen<strong>in</strong>g flexibility - Screen<strong>in</strong>g parameters can be selected and/or modified<br />

based on customer requirements. Level 1 and level 2 can be carried out on<br />

additional TLRs, <strong>in</strong>clud<strong>in</strong>g mouse TLRs. More compound concentrations can be<br />

tested.<br />

Cost effective - A set-up charge applies for the first compound. Subsequent<br />

compounds are heavily discounted.<br />

Description<br />

TLR Ligand Screen<strong>in</strong>g<br />

Two choices of services are offered, level 1 and level 2, that can be performed<br />

sequentially or <strong>in</strong>dividually.<br />

Level 1: S<strong>in</strong>gle dose test<strong>in</strong>g on a panel of human TLRs<br />

Test<strong>in</strong>g is carried out on seven TLRs.<br />

- TLR2: wide array of microbial molecules<br />

- TLR3: dsRNA<br />

- TLR4: Gram-negative lipopolysaccharide<br />

- TLR5: flagell<strong>in</strong><br />

- TLR7: s<strong>in</strong>gle-stranded RNA and small antiviral molecules<br />

- TLR8: s<strong>in</strong>gle-stranded RNA and small antiviral molecules<br />

- TLR9: specific unmethylated CpG oligonucleotide sequences<br />

Screen<strong>in</strong>g is performed at a s<strong>in</strong>gle concentration, typically a 1/10 dilution of the<br />

orig<strong>in</strong>al compound solution provided, or customer specified. Duplicate tests (plus<br />

controls, see list).<br />

Level 2: Dose response on one or two TLRs<br />

Three concentrations of the compound(s), typically 1/10, 1/100 and 1/1000<br />

dilutions of the orig<strong>in</strong>al compound solution, are tested on the TLR(s) recogniz<strong>in</strong>g<br />

the compound(s) as determ<strong>in</strong>ed <strong>in</strong> level 1 or specified by the customer. Triplicate<br />

tests (plus controls).<br />

TLR ligands are typically recognized by a s<strong>in</strong>gle TLR, potentially two (TLR7 and<br />

TLR8). Recognition by TLR4 usually reflects the presence of endotox<strong>in</strong>s <strong>in</strong> the<br />

sample.<br />

NOD Ligand Screen<strong>in</strong>g<br />

Two cytoplasmic prote<strong>in</strong>s <strong>in</strong>volved <strong>in</strong> pathogen recognition, NOD1 and<br />

NOD2, can be added to this screen<strong>in</strong>g service. Their activation by a given<br />

compound is detected us<strong>in</strong>g the same method as the one described above.<br />

Compounds can be supplied as stock solutions or as solid materials for watersoluble<br />

compounds.<br />

A detailed report will be prepared and provided to the customer electronically<br />

and <strong>in</strong> hard copy. All procedures are performed accord<strong>in</strong>gly to strict guidel<strong>in</strong>es.<br />

Confidentiality is guaranteed.<br />
