Hygromyc<strong>in</strong> B<br />
Description<br />
Hygromyc<strong>in</strong> B is an am<strong>in</strong>oglycoside antibiotic produced by Streptomyces<br />
hygroscopicus. It <strong>in</strong>hibits prote<strong>in</strong> synthesis by <strong>in</strong>terfer<strong>in</strong>g with translocation<br />
and caus<strong>in</strong>g mistranslation at the 70S ribosome. Hygromyc<strong>in</strong> B is effective<br />
on most bacteria, fungi and higher eukaryotes. Resistance to hygromyc<strong>in</strong> is<br />
conferred by the hph gene from E. coli. Hygromyc<strong>in</strong> B is normally used at<br />
a concentration of 50-200 μg/ml <strong>in</strong> mammalian cells and 100 μg/ml <strong>in</strong><br />
bacteria.<br />
Two grades of Hygromyc<strong>in</strong> B are available:<br />
• Hygromyc<strong>in</strong> B (purity >85%)<br />
• HygroGold (purity >98%)<br />
Phleomyc<strong>in</strong><br />
Description<br />
Phleomyc<strong>in</strong> is a glycopeptide antibiotic of the bleomyc<strong>in</strong> family, isolated<br />
from a mutant stra<strong>in</strong> of Streptomyces verticillus. It b<strong>in</strong>ds and <strong>in</strong>tercalates<br />
DNA thus destroy<strong>in</strong>g the <strong>in</strong>tegrity of the double helix. Phleomyc<strong>in</strong> is active<br />
aga<strong>in</strong>st most bacteria, filamentous fungi, yeast, plant and animal cells. Use<br />
of phleomyc<strong>in</strong> is recommended for cells poorly sensitive to Zeoc<strong>in</strong> , i.e.<br />
filamentous fungi and some yeasts. Phleomyc<strong>in</strong> resistance is conferred by<br />
the Sh ble gene from Streptoalloteichus h<strong>in</strong>dustanus which encodes a prote<strong>in</strong><br />
that b<strong>in</strong>ds to phleomyc<strong>in</strong>, <strong>in</strong>hibit<strong>in</strong>g its DNA cleavage activity. Typically,<br />
phleomyc<strong>in</strong> is used at a concentration of 10 μg/ml for yeasts and 25-150<br />
μg/ml for filamentous fungi.<br />
Zeoc<strong>in</strong> <br />
Description<br />
Zeoc<strong>in</strong> is a formulation of phleomyc<strong>in</strong> D1, a copper-chelated<br />
glycopeptide antibiotic produced by Streptomyces CL990. Zeoc<strong>in</strong> causes<br />
cell death by <strong>in</strong>tercalat<strong>in</strong>g <strong>in</strong>to DNA and cleav<strong>in</strong>g it. This antibiotic is<br />
effective on most aerobic cells and is therefore useful for selection <strong>in</strong> bacteria,<br />
eukaryotic microorganisms, plant and animal cells. Resistance to Zeoc<strong>in</strong> is<br />
conferred by the Sh ble gene product which <strong>in</strong>activates Zeoc<strong>in</strong> by b<strong>in</strong>d<strong>in</strong>g<br />
to the antibiotic. Zeoc<strong>in</strong> is used at a concentration of 50-300 μg/ml for<br />
selection <strong>in</strong> mammalian cells and 25 μg/ml for bacterial selection.<br />
Contents and Storage<br />
Hygromyc<strong>in</strong> B and HygroGold are provided as 100 mg/ml yellow<br />
solutions. HygroGold is also provided as a powder. Products are shipped<br />
at room temperature. Store at -20°C. Hygromyc<strong>in</strong> B solutions are stable<br />
for at least one year when stored at -20°C.<br />

Phleomyc<strong>in</strong> is shipped at room temperature. Store the solution at -20°C<br />
and the powder at 4°C. Phleomyc<strong>in</strong> is stable for at least one year when<br />
properly stored.<br />
