The objectives of this study will be to 1) survey potential six-rowed malting barley parents for variation of candidate marker alleles to key malting barley DP QTL, 2) concurrently conduct conventional and marker-assisted selection for desirable malting quality in a good agronomic background, and 3) compare marked donor regions as to their contributions toward enhanced malting quality. It is anticipated that the information and germplasm obtained from this work will lead to the development of new and improved six-rowed malting barley cultivars for the Western intermountain region of the U.S. The initial emphasis will be placed on DP QTL polymorphisms and polymorphisms among Colter and the six-rowed malting standard 'Morex'. METHODS Genomic DNA was extracted from the above cultivars using a miniprep procedure that was adapted from the DNA extraction protocol given in the 1994 ITMI Wheat Mapping Workshop Laboratory Manual pp. 19-21. In addition to Colter and Morex, the mapping parent 'Steptoe' will be added to the survey gels as a control to to validate mapped bands. Techniques for PCRRAPD followed that of Hoffman and Bregitzer (1996); for AFLP, that of Hoffman et al. (2000); and for PCR-STS, that of Tragoonrung et al. (1992). PROGRESS TO DATE Marker Analyses In 2000, we reported on the polymorphism pattern of PCR-RAPD marker AB11M950 that maps near RFLP locus VATP57A which is in between Brz and Amy2, a region where DP QTL is found along with several of other malting and agronomic QTL. Last year, a similar, but not identical polymorphism pattern to AB11, was found for the lower molecular weight band generated by the PCR-STS primer pair KV1, KV9. This primer pair was designed in Dr. Tom Blake's laboratory (Tragoonrung et al., 1992) from sequences of B1-Hordein encoded by Hor2 (Shewry et al., 1988) which maps to the short arm of barley chromosome 5(1H) where a DP QTL was identified by Hayes et al. (1993). No other major malting or agronomic QTL have associated with this chromosomal region. So far, none of the AFLP markers that mapped in DP QTL regions have yielded useful polymorphism patterns (polymorphic between Morex and Colter and between Morex and Steptoe, and band-positive for Morex) as described for AB11M950 (chromosome 1) and KV1, KV9 (chromosome 5). Crosses Made and Segregants Tested The first cross to be made for marker analysis was Colter/Morex. An F1 from this cross was backcrossed several times to Colter. The idea is to extract malting quality QTLs such as DP from Morex and place these in an adapted background such as Colter. The resulting BC1F1s were tested with the KV1, KV9 primers which amplify sequences in Hor2 on the short arm of chromosome 5. Both phenotypic and marker selection is to be employed. The BC1F1s with the Morex band were backcrossed again to Colter and screened for KV1, KV9 segregation. The resulting BC2F1s were again screened for KV1, KV9 segregation and ones with Morex alleles were selected for selfing and further
33 backcrossing. The selfed lines will be selected for the Morex allele and advanced to the BC2F2:3 generation for initial field selection and testing of DP. This first BC population did not segregate for chromosome 1 marker AB11M950. It is thought that some intravariety variability is present in Morex. A new parent from the mapping stock was chosen to derive a new backcross series. The new series is now in the BC1 generation and will allow selection with KV1, KV9 and AB11M950. This study has been expanded to include additional 6-rowed varieties and germplasms for tests of DP QTL in different backgrounds across several environments. This expanded version will include the Bmy1 region of chromosome 4 and has received one-year funding from the North American Barley Genome Project. REFERENCES Foster, A E., G.A.Peterson, and O.J.
