Description: These vectors are designed specifically for high-level expression<br />

of proteins in mammalian cells from the CMV promoter with or without a<br />

selectable marker. The pFN9A Vector provides Renilla luciferase, which may be<br />

used as a transfection control. The pFN9A Vector was designed to complement<br />

pGL4 firefly luciferase vectors when exogenous proteins (e.g., a receptor<br />

of transcription factor) must be expressed for reporter assays. All inserts may<br />

be confirmed by cell-free expression with the TNT ® T7 Quick System (Cat.#<br />

L1170).<br />

Note: Flexi ® Vectors contain the lethal barnase gene to reduce background<br />

colonies without inserts during the subcloning procedure. Using the Flexi ® Vector<br />

Cloning System replaces the barnase gene with your insert. These vectors,<br />

as purchased, cannot be cultured in normal laboratory strains of E. coli without<br />

an insert.<br />

Description: These vectors are designed specifically for expression of proteins<br />

in E. coli from the T7 RNA polymerase promoter with or without the addition<br />

of purification tags. Expression levels can vary from protein to protein, but in<br />

general the N-terminal HaloTag ® fusion protein (e.g., pFN18A/K) can increase<br />

expression by increasing solubility of the expressed protein. Proteins may be<br />

purified using the HaloTag ® fusion with the HaloTag ® Protein Purification System<br />

(Cat.# G6280). Expression may be confirmed through cell-free expression<br />

with the S30 T7 High-Yield Protein Expression System (Cat.# L1110).<br />

The Flexi ® Vector System is a simple, yet powerful, directional cloning method<br />

for protein-coding sequences. It is based on two rare-cutting restriction<br />

enzymes, SgfI and PmeI, and provides a rapid, efficient and high-fidelity way to<br />

transfer protein-coding regions between a variety of Flexi ® Vectors without the<br />

need to resequence.<br />
