Description: The MagneGST Protein Purification System provides a simple,<br />

rapid and reliable method for the purification of glutathione-S-transferase (GST)<br />

fusion proteins. Immobilized glutathione paramagnetic particles (MagneGST<br />

Particles) are used to isolate GST-tagged protein directly from a crude or<br />

cleared lysate using either a manual or automated procedure and requires use<br />

of a magnetic stand. GST-tagged proteins can be purified on a small scale from<br />

1ml of culture or on a large scale using more than 50ml of culture. Samples<br />

also can be processed using a robotic platform.<br />

• Quick: After cell lysis, no requirement for high-speed centrifugation to<br />

clear lysate.<br />

• Scalable: Scalable protocol using 1–50ml of cell culture.<br />

Storage Conditions: The complete system consists of two individual parts,<br />

Description: The MagneGST Pull-Down System is designed for detection of<br />

protein interactions between GST-fusion proteins expressed in bacterial lysates<br />

and prey proteins expressed in the TnT ® Systems. Prey protein synthesized<br />

in the TnT ® Quick Coupled Transcription/Translation Reaction is captured<br />

using bait protein (GST-fusion protein) immobilized on MagneGST Particles.<br />

Nonspecifically bound proteins are then washed away, and the prey protein is<br />

analyzed. Prey proteins can be detected by incorporating radioactively labeled<br />

methionine in the TnT ® Quick reaction, followed by SDS-PAGE and autoradiography<br />

or by incorporating the supplied non-radioactive methionine in the TnT ®<br />

reaction and detecting by Western blotting with protein-specific antibodies.<br />

Storage Conditions: Store the TnT ® T7 Quick Master Mix and Methionine at<br />

–70°C. Store the RQ1 RNase-Free DNase at –20°C. Store the Nuclease-Free<br />

Water, MagneGST Glutathione Particles, MagneGST Binding/Wash Buffer<br />

and Cell Lysis Reagent at 4°C.<br />

Description: The Maxwell ® 16 Polyhistidine Protein Purification Kit is used<br />

with the Maxwell ® 16 Instrument to provide an easy method for the efficient,<br />

automated purification of polyhistidine-tagged protein from bacterial cultures<br />

and other sample types including mammalian and insect cells. With minor<br />

modifications, the reagents can also be used for purification of HQ-tagged<br />

proteins from bacterial cultures.<br />

The Maxwell ® Instrument is supplied with a preprogrammed purification<br />

procedure and reagent cartridges specifically designed to maximize<br />

simplicity and convenience. The instrument can process up to 16 samples in<br />

approximately 40 minutes. The purified protein is compatible with downstream<br />

applications such as gel electrophoresis and Western blot analysis.<br />
