Description: Pfu DNA Polymerase is a thermostable enzyme of approximately<br />

90kDa isolated from Pyrococcus furiosus. The enzyme replicates DNA at 75°C,<br />

catalyzing the polymerization of nucleotides into duplex DNA in the 5´→3´<br />

direction in the presence of magnesium. Pfu DNA Polymerase also possesses<br />

3´→5´ exonuclease (proofreading) activity. Base misinsertions that may occur<br />

during polymerization are rapidly excised by the proofreading activity of the<br />

polymerase. Consequently, Pfu DNA Polymerase is recommended for use in<br />

PCR and primer extension reactions that require high-fidelity synthesis. Pfu<br />

DNA Polymerase-generated PCR fragments are blunt-ended.<br />

Pfu DNA Polymerase 10X Reaction Buffer with MgSO 4 : 200mM Tris-HCl<br />

(pH 8.8 at 25°C), 100mM KCl, 100mM (NH 4 ) 2 SO 4 , 20mM MgSO 4 , 1.0% Triton ®<br />

X-100 and 1mg/ml nuclease-free BSA.<br />

Description: High-quality deoxynucleotide triphosphates (dNTPs) are critical for<br />

PCR success. The PCR Nucleotide Mix is a premixed solution containing sodium<br />

salts of dATP, dCTP, dGTP and dTTP, each at a concentration of 10mM in water<br />

at pH 7.5; the total concentration of nucleotides is 40mM. This solution is ready<br />

to use and is optimized for standard PCR and specialty approaches including<br />

hot-start and reverse transcription PCR (RT-PCR). One microliter (1μl) is sufficient<br />

for amplification in a typical 50μl reaction volume.<br />
