Thema 3: Auswirkungen auf Pflanzen, Tiere und Ökosystemecal features such as fibre frequency, fibre lumina, thickness ofvessel walls, cell wall area were unaffected by precipitation. Lowprecipitation was furthermore related to a remarkable increase inthe latewood δ13C of beech trees (increase of isotope 13 C concentration). To show the importance of tree nutritions for wood formation aminoacid contents in cambium tissues were measured. Aspartic, glutamic and asparagine acids are dominated in the amino – Npool of cambium tissues of beech wood during June and August. The asparagine content, a transport compound of nitrogen, decreasedfrom June to August, whereas both glutamic aand asparticacids levels were unaffected by seasonal changes. To get insight into the molecular background of wood formationduring the course of a year, we performed RNA sequencing (RNA-Seq) of samples from the above described site Calvörde: cambiumtissue of four harvests during the year 2010 (April, June, Augustand October) was analyzed. Differentially expressed genes relatedto wood formation have been chosen from this data and primerswere designed. Those genes have been then tested by quantitativeReal-time PCR during the course of several years (2009-2012) andon the different sites. In summary, years or sites with low precipitation result in changesof some anatomical features (radial growth rate, vessel numberand vessel lumina) which can be related to tree nutrition (carbonisotope composition (δ13C) and amino acid compounds) as well asto gene expression of beech trees.
