<strong>The</strong> first figure shows the connections from the mouthpiece in the direct system to the NO analyser,<br />

the CO2 analyser, the pressure analyser and the chart recorder. <strong>The</strong> second figure shows the t-piece<br />

connection allowing additional measurement <strong>of</strong> flow.<br />

6.5.3 Ethics and Consent<br />

<strong>The</strong> studies received prospective approval by the Royal Brompton Hospital Ethics Committee<br />

and informed consent was obtained from each <strong>of</strong> the volunteer subjects.<br />

6.5.4 <strong>The</strong> subjects<br />

In total, twenty healthy volunteers (mean age 35.9 years, range 18-52 years, 9 males) took<br />

part in these methodological studies. Following consent, each subject completed a<br />

questionnaire (see Appendix 2). <strong>The</strong> subjects were non-atopic with no history <strong>of</strong> respiratory<br />

or cardiac disease and taking no medication. All had normal spirometry (Compact<br />

Vitalograph, Vitalograph Ltd, Buckingham, United Kingdom). Two female subjects involved<br />

in the variable flow and variable pressure experiments smoked two cigarettes per day; the rest<br />

were nonsmokers.<br />

6.5.5 Statistical analvsis<br />

<strong>The</strong> results were analysed using the Statistical Products and Services Solutions, (Package U<br />

7.0, SPSS Inc, Chicago, USA). This employs the Student's t-test for matched pairs with mean,<br />
