ABSTRACT<br />

We previously reported that storing mobilized peripheral <strong>blood</strong> progenitor cell<br />

(PBPC) collections overnight before CD34 +<br />

selection using the CellPro CEPRATE SC<br />

Stem Cell Concentration System may be associated with delayed platelet count<br />

recovery after high-dose chemotherapy <strong>and</strong> reinfusion (Koc ON, Gerson SL, Phillips<br />

GL, Cooper BW, Kutteh L, Van Zant G, Reece DE, Fox RM, Schupp JE, Tainer N,<br />

Lazarus HM: Autologous CD34 +<br />

cell <strong>transplantation</strong> for patients with advanced<br />

lymphoma: Effects of overnight storage on peripheral <strong>blood</strong> progenitor cell enrichment<br />

<strong>and</strong> engraftment. Bone Marrow Transplant 21:337-343, 1998). To investigate this<br />

issue, we undertook a r<strong>and</strong>omized, three-center trial comparing overnight storage prior<br />

to CD34 selection (at 4°C) vs. immediate CD34 selection <strong>and</strong> cryopreservation of<br />

PBPC collections in 68 lymphoma patients undergoing <strong>autologous</strong> transplant. PBPCs<br />

were mobilized with cyclophosphamide 4 g/m 2<br />

intravenously, prednisone 2 mg/kg/d<br />

orally for 4 days, <strong>and</strong> granulocyte colony-stimulating factor (G-CSF) 10 pg/kg/d<br />

subcutaneously. Tliirty-four patients were r<strong>and</strong>omized to overnight storage while 34<br />

patients were assigned to immediate processing. Fifteen patients were excluded from<br />

the primary analysis of time to platelet recovery: three had tumor progression before<br />

autotransplant <strong>and</strong> 12 received unselected cells alone or in combination with selected<br />

cells since
