selection to support multiple cycles of such therapy has not been examined.<br />

To date, we have treated 42 patients with metastatic or locally advanced breast<br />

cancer as part of phase I or II studies using three cycles of high-dose chemotherapy<br />

delivered every 28 days, with each cycle supported by PBPCs <strong>and</strong> G-CSF. The<br />

primary purpose of these studies was to develop a suitable outpatient regimen for<br />

a subsequent phase III study. The results of these studies are presented here.<br />

METHODS<br />

Eligibility criteria<br />

Patients with histologically proven metastatic or locally advanced breast cancer<br />

were prospectively entered into four consecutive studies using three cycles of high-<br />

dose chemotherapy with each cycle being supported with PBPCs <strong>and</strong> G-CSF<br />

(Table 1). Sufficient PBPCs to support these three cycles of high-dose therapy were<br />

collected before the first cycle of high-dose therapy. Patients were excluded if they<br />

had significant cardiac, hepatic, or renal impairment, an Eastern Cooperative<br />

Oncology Group (ECOG) performance status >2, ANC
