We have shown that the RES complexhas the ability to modulate alternative splicing of artificial reporters. Correlation ofthe structure and function indicates that the probable phosphothreonine-bindingpocket of Pml1 is not involved in this alternative splicing control. This work waspublished (Structure of the yeast Pml1 splicing factor and its integration into the REScomplex. Brooks MA, Dziembowski A, Quevillon-Cheruel S, Henriot V, Faux C,van Tilbeurgh H, Séraphin B.Nucleic Acids Research 2009 37(1):129-143). We arecurrently trying to define the molecular function of the RES complex during splicingand to identify its partner(s).• WP10- Analysis of the structure of the Pml1 subunit of the RES complex revealeda probable phosphothreonine-binding pocket. We have tried to identify partnersbinding to this site. Functional analysis has, however, demonstrated that a functionalphosphothreonine-binding pocket is not required to mediate the alternative splice siteactivity of the RES complex.• WP13- A collaborative manuscript on the role of the various domains of Mud2 isin preparation with partner 1b.- The RES complex has been shown to affect pre-mRNA leakage and is thuspotentially involved in the early steps of intron recognition. We are currently tryingto define the molecular function of the RES complex and how it affects pre-mRNAleakage.• WP15- We have used the RES complex as a test case to develop a mutagenesisstrategy to map protein interaction surfaces. This method was successful and it isreported in our publication describing a structure/function analysis of this assembly(Structure of the yeast Pml1 splicing factor and its integration into the RES complex.Brooks MA, Dziembowski A, Quevillon-Cheruel S, Henriot V, Faux C, vanTilbeurgh H, Séraphin B.Nucleic Acids Research 2009 37(1):129-143).• WP18- Bertrand Séraphin participated to the mentor lunch in Krackow(Deliverable 239).157
