Participant 20 – Alberto Kornblihtta) Work performed during the period• WP13Our group continued investigating the mechanisms that couple the transcription and splicingmachineries. We payed particular attention to the physiological and pathological signals that affectalternative splicing decisions through the mentioned coupling mechanisms. For example, we found thatin neuronal cells, membrane depolarization, an indicator of nueronal activity, affects alternativesplicing of the NCAM mRNA by modifying the chromatin structure of its gene in a way that favorshigher transcription elongation rates that in turn decrease splicing of the alternative exon 18, involvedin the regulation of neuron differentiation. On the other hand we studied how DNA damage caused byUV irradiation affects alternative splicing of many genes involved in apoptosis and cell survival. Wefound that UV irradiation triggers a mechanism that hyperphosphorylates the carboxy terminal domainof RNA polymerase II, promoting a reduction in pol II elongation, which in turn affects alternativesplicing.• WP17During <strong>2008</strong> we intensified collaborations within <strong>EURASNET</strong>. The graduate student Ezequiel Petrillovisited the labs of Andrea Barta in Vienna and of John Brown in Dundee to study the regulation ofalternative splicing in plants by the light.
