IMMUNOACTIVATION METHOD FOR SPECIFIC
DETERMINATION OF PMN-ELASTASE IN PLASMA
S. Domazetovska, M. Bogdanova
Institute of Clinical Biochemistry, Medical Faculty, Skopje, Macedonia
PMN-elastase is a neutral proteinase from lysosomes of polymorphonuclear
leucocytes. The release of elastase in circulation is one of
the results of inflammatory response. A sensitive, specific and fast
immunoassay method to determine the extent of release of elastase
was developed by Merck. It is an immunoactivation method.
Material and method: Antibody fragments against human PMNelastase
are covalently bound to peroxidase and produce antibody conjugates.
The antibody conjugate and the PMN-elastase in the sample
form aggregates. In non-aggregated antibody conjugates, the peroxidase
is inhibited. In the aggregates the peroxidase catalyses the oxidation
of 4-aminophenasone to quinonimine dye. EDTA plasma
samples from 40 healthy adults, 41 samples from newborns, were
analysed using Merck test for immunoassay and Photometer Vitalab
Microlab Merck at 500 nm, 37°C.
