DETERMINATION OF PMN-ELASTASE IN PLASMA<br />

PMN-elastase is a neutral proteinase from lysosomes of polymorphonuclear<br />
leucocytes. The release of elastase in circulation is one of<br />
the results of inflammatory response. A sensitive, specific and fast<br />
immunoassay method to determine the extent of release of elastase<br />
was developed by Merck. It is an immunoactivation method.<br />
Material and method: Antibody fragments against human PMNelastase<br />
are covalently bound to peroxidase and produce antibody conjugates.<br />
The antibody conjugate and the PMN-elastase in the sample<br />
form aggregates. In non-aggregated antibody conjugates, the peroxidase<br />
is inhibited. In the aggregates the peroxidase catalyses the oxidation<br />
of 4-aminophenasone to quinonimine dye. EDTA plasma<br />
samples from 40 healthy adults, 41 samples from newborns, were<br />
analysed using Merck test for immunoassay and Photometer Vitalab<br />
Microlab Merck at 500 nm, 37°C.<br />
