The physiological functions of these systems are analyzed by theproduction and analysis of transgenic and gene-targeted animal models. Among the hormonesstudied, serotonin is of special interest, since it is not only involved in vascular homeostasis andother peripheral functions, but also serves as potent and multifunctional neurotransmitter inthe brain. In addition, the group is interested in embryology and stem cell research, aiming toapply these fields to the rat and to establish gene targeting in this species.Renin-angiotensin systemNatalia Alenina, Brit Rentzsch, Mihail Todiras, Ping Xu,Aline Hilzendeger, Luiza Rabelo, Sergio Santos, PhilipBoyé, Gabin Sihn, Cibele CardosoThe renin-angiotensin system (RAS) is of central importancein blood pressure regulation and in the initiationof target organ damage. In particular, local angiotensin-II generating systems in tissues such as brain, heart,vessels, and kidney are involved in these processes.Therefore, transgenic rats with local up- or downregulationof RAS components in these organs, e.g. by the localexpression of antisense-RNA or of a peptide-liberatingprotein, were produced and analyzed to clarify the localfunctions of angiotensin II. Other genetically alteredmouse and rat models for non-classical RAS componentssuch as ACE2, the renin receptor, angiotensin(1-7)and its receptor Mas, have elucidated the physiologicalfunction of these factors. Together with transgenic ratsoverexpressing this peptide, Mas-knockout mice characterizedthe angiotensin(1-7)/Mas system as a cardioprotectiveaxis that counteracts the classical RASeffects in particular improving endothelial function.Furthermore, these animals showed that angiotensin(1-7) and Mas are important for insulin sensitivity andthe pathogenesis of metabolic syndrome.Kallikrein-kinin systemInes Schadock, Robert Fischer, Marcelo Mori, CarlosBarros, Alessander Guimaraes, Fatimunnisa Qadri,Anthony Rousselle, Johan Duchene, Vanessa MerinoThe kallikrein-kinin system (KKS) is an important hormonesystem for cardiovascular regulation also mostlycounteracting the effects of the RAS. As models for thefunctional analysis of the KKS in intact animals, transgenicrats were generated expressing different componentsof the system, such as tissue kallikrein, the kininB1 or the B2 receptor either ubiquitously or specificallyin cardiovascular organs. These animals supported theprotective role of the KKS in kidney and heart againstischemic, diabetic, and hypertrophic injury. Knockoutmice for the kinin B1 receptor were generated andrevealed important functions of this protein in painperception and inflammation. Moreover, the B1 receptorturned out to be involved in sepsis, stroke, multiple sclerosisand high-fat diet induced obesity. Mice lackingboth kinin receptors and thereby being devoid of afunctional KKS were also generated and shown to becompletely normal at baseline suggesting that the KKSis irrelevant for development and basic regulation ofthe cardiovascular system but is involved in the pathogenesisof multiple diseases and, thus, a relevant drugtarget.48 Cardiovascular and Metabolic Disease <strong>Research</strong>
