The test material was 94.5% chemically and 98.25% radiochemically pure active <strong>in</strong>gredient<br />

and had a specific activity of 1.058 GBq/mmol (batch No. of labelled compound, TSN 102298, DE3-<br />

E1004-77, INV1893).<br />

Renal excretion was the major route of elim<strong>in</strong>ation <strong>in</strong> all three groups. In the group of pregnant<br />

rabbits that were pre-treated with nonlabelled am<strong>in</strong>opyralid, faecal excretion was substantially<br />

less, while renal excretion was m<strong>in</strong>imally <strong>in</strong>creased when compared with that <strong>in</strong> the other groups.<br />

The only radioactive peak <strong>in</strong> pooled samples of ur<strong>in</strong>e and faeces was identified as unchanged<br />

a m<strong>in</strong>opyralid.<br />

Other than the gastro<strong>in</strong>test<strong>in</strong>al tract, which showed some radioactivity at term<strong>in</strong>ation, the<br />

spleen <strong>in</strong> the group of pregnant rabbits that were pre-treated with nonlabelled am<strong>in</strong>opyralid was the<br />

only organ show<strong>in</strong>g some radioactivity.<br />

In all dosed groups, radioactivity was detected <strong>in</strong> the plasma with<strong>in</strong> 10 m<strong>in</strong> after dos<strong>in</strong>g and<br />

T max<br />

was achieved with<strong>in</strong> 1 h. The plasma elim<strong>in</strong>ation half-life was lowest <strong>in</strong> the group of pretreated<br />

pregnant rabbits (4.1 h) and highest <strong>in</strong> the group of non-pregnant rabbits (7.2 h). There was <strong>in</strong>creased<br />

early renal excretion, shorter T max<br />

and higher plasma area under the curve of concentration–time<br />

(AUC) <strong>in</strong> pregnant rabbits given repeated doses, <strong>in</strong>dicat<strong>in</strong>g more rapid and extensive absorption <strong>in</strong><br />

these animals when compared with non-pregnant rabbit or pregnant rabbits given a s<strong>in</strong>gle dose on<br />

day 7 of gestation.<br />
