of allergic reaction to a formulated product. There were no reports of poison<strong>in</strong>g and no reports of<br />

s ensitization dur<strong>in</strong>g use of the formulated product.<br />

The Meet<strong>in</strong>g concluded that the exist<strong>in</strong>g database was adequate to characterize the potential<br />

hazards to fetuses, <strong>in</strong>fants and children.<br />

<strong>Toxicological</strong> evaluation<br />

An acceptable daily <strong>in</strong>take (ADI) of 0–0.01 mg/kg bw was established for difenoconazole<br />

based on the NOAEL of 1.0 mg/kg bw per day <strong>in</strong> rats, identified on the basis of reduced body-weight<br />

ga<strong>in</strong>s, reduced platelet counts and hepatic hypertrophy <strong>in</strong> a 24-month long-term dietary study of<br />

toxicity and carc<strong>in</strong>ogenicity. A safety factor of 100 was applied. The <strong>in</strong>creased <strong>in</strong>cidence of hepatocellular<br />

adenomas and carc<strong>in</strong>omas observed <strong>in</strong> mice at 423 mg/kg bw per day, the NOAEL be<strong>in</strong>g 4.7<br />

mg/kg bw per day, <strong>in</strong> an 18-month long-term dietary study of toxicity and carc<strong>in</strong>ogenicity, was likely<br />

to be due to a mode of action without human relevance at exposure levels that do not cause changes<br />

<strong>in</strong> the liver.<br />

An acute reference dose (ARfD) of 0.3 mg/kg bw was established for difenoconazole. This was<br />

based on the NOAEL of 25.0 mg/kg bw <strong>in</strong> rats, identified on the basis of cl<strong>in</strong>ical signs <strong>in</strong> a s<strong>in</strong>gledose<br />

study of neurotoxicity and us<strong>in</strong>g a safety factor of 100. This ARfD is supported by the NOAEL<br />

of 25 mg/kg bw per day for maternal toxicity <strong>in</strong> a study of developmental toxicity <strong>in</strong> rats and rabbits<br />

on the basis of excess salivation <strong>in</strong> rats at 100 mg/kg bw per day and body-weight loss <strong>in</strong> rabbits<br />

d ur<strong>in</strong>g the first few days of treatment at 75 mg/kg bw per day.<br />
