Antimetabolites
Cost : Inj: 1 g vial (Rs. 322.20)
Dacarbazine
I : Malignant melanoma, Hodgkins, Soft tissue sarcomas
C/I : Pregnancy, lactation, persons with severe
myelosuppression.
P/C : Hepatic / renal impairment, haematologic monitoring,
restrict food intake 4-6 hrs before therapy.
A/E : Nausea, vomiting, diarrhoea, anaphylaxis, bone marrow
depression, urticaria, photosensitivity, hepatic necrosis,
renal impairment, thrombocytopenia, blurred vision,
flu-like syndrome, myalgia.
P/A : Injection 200mg vial
Dose : 2.5 - 4.5 mg/kg/day x 5-10 days or 250 mg/m 2 /day x 5
days every 3 weeks or 850mg/ m 2 every 3-6 weeks
given IV.
D/ I : lmpairs the immunogenicity of the live attenuated
vaccine. It forms precipitate with the hydrocortisone
hemisuccinate.

Temozolamide :
I: Glioblastoma, Anaplastic Astrocytoma, refractory to
nitrosoureas, Melanoma, and Metastatic Carcinomas
to brain.
C/I : Hypersensitivity to dacarbazine
A/E : Myelosuppression, Nausea, Vomiting, Alopecia
Dosage
Schedule: 1. 150 to 200 mg/m 2 orally on an empty stomach
for 5 days every 28 days.
2. 75 mg/ m 2 orally on an empty stomach daily
during radiation therapy for up to 7 weeks.
