loop), and a thumb (the F helix). This metal-b<strong>in</strong>d<strong>in</strong>g motif has a strong aff<strong>in</strong>ity<br />

for calcium ions. The motif is generally consi<strong>de</strong>red as an octahedral arrangement<br />

around the calcium ion with one site shared by both carboxylate oxygen<br />

atoms of a glutamate si<strong>de</strong> cha<strong>in</strong> to give a coord<strong>in</strong>ation number of 7. The aff<strong>in</strong>ity<br />

of this arrangement for calcium ions is highest when there is a pair of contiguous<br />

EF hands. For each hand the aff<strong>in</strong>ity is related to the net ligand charge of<br />

the loop, and the hydrophobicity of various residues <strong>in</strong> the EF-hand pair [76].<br />

Nature has utilized this calcium-specific b<strong>in</strong>d<strong>in</strong>g motif <strong>in</strong> several prote<strong>in</strong>s, such<br />

as tropon<strong>in</strong> C, calmodul<strong>in</strong>, parvalbum<strong>in</strong>, and lactalbum<strong>in</strong>.
