4.8 Biotransformation and Kinetics<br />

Type: animal<br />

Method: Oysters and clams were collected from Lake Pontchartrain,<br />

Louisiana, USA in May and June 1980 and analysed for the<br />

presence of benzene.<br />

Result: One sample of oysters (Crassostrea virginica) was found to<br />

contain 220 µg/kg wet weight and one sample of clams (Rangia<br />

cuneata) contained 260 µg/kg wet weight. A second sample of<br />

clams taken from the same area had no detectable benzene<br />

(detection limits not stated).<br />

Method: The effects of benzene on various mixed–function oxydases<br />

and on cytochrome P450 was investigated.<br />

Remark: Significant methodological deficiencies<br />

No controls treated with vehicle (corn oil) only were<br />

reported to be used.<br />

Result: Cytochrome P450 content was decreased after benzene<br />

treatment to ca. 78% of control. Among the monooxygenase<br />

dependent enzymes studied, the activity of the 7–ethoxy<br />

resorufin–O–deethylase (EROD) was decreased (73% of<br />

control). The activity of the aniline 4–hydroxylase and<br />

ethylmorphine N–demethylase was not affected.<br />

Test condition: Gilthead seabream (Sparus aurata. weight 225–275 g) were<br />

kept in aerated, filtered seawater at 16.5–17.5 degree C and<br />

fed daily. Benzene was administered once daily for 4 days<br />

by s.c injection (0.3 ml of benzene/corn oil 1:1).<br />

Microsomes were prepared from the liver and the cytochrome<br />

P450 content and the activity of several monooxygenases were<br />

determined by standard methods.<br />

Controls receiving corn oil are not reported.<br />

Type: aquatic<br />

Method: Between November 1980 and August 1981, quarterly, five<br />

replicate, one week composites of the final effluent from<br />

the Los Angeles County wastewater treatment plant were<br />

collected for analysis of extractable organics (EOs) and two<br />

grab samples of surficial sediments were collected for<br />

analysis of volatile organics (VOs). The sampling site was<br />

6 km NW of the discharge zone at Pulos Verdes and a modified<br />

Van Veen sampling device was used. During June 1981,<br />

biological samples were collected at this same site. These<br />

included Citharichthys xanthostigma (Pacific sanddab),<br />

Microstomus pacificus (Dover sole), Scorpaena guttata<br />

(scorpionfish), Genyonemus lineatus (white croaker),<br />

Sicyonia ingentis (ridgeback prawn) and a sample of small<br />
