______________________________________________________________________________
Type: Cytogenetic assay
Species: rat Sex: male
Strain: Wistar
Route of admin.: i.p.
Exposure period: single dose
Doses: 878 mg/kg bw
Method: other
GLP: no data
Test substance: other TS
Remark: Groups of five male Wistar–derived, outbred, Alderely Park
rats were injected with benzene or, in the case of the
control group, water. The animals were killed 24 hours
later, the bone marrow samples prepared according to the
method of Sugiyama T. J. natn. Cancer Inst. 47, 1267–1275,
1971 but with slight modifications and the slides examined
for any chromosome abnormalities.
Reliability: 2 (valid with restriction)
Comparable to guideline study with acceptable restrictions
no positive control, one sampling time, one sex, one dose
Result: Benzene caused a statistically significant increase in the
numbers of cells with abnormalities and chromatid and
chromosome gaps and breaks.
Test substance: Laboratory reagent grade; purity not specified.
Type: Cytogenetic assay
Species: rat Sex: male
Strain: Wistar
Route of admin.: inhalation
Exposure period: a) single exposure for 2 h , b) 6 h per day for 5 days
Doses: 0, 750 or 7500 ppm (2.4 or 24 mg/l)
Method: other
GLP: no data
Test substance: other TS
Remark: Groups of at least 7 male Wistar–derived, outbred, Alderely
Park rats were exposed to benzene or, in the case of the
control group, to air. The animals were killed 6 (b) or 24
(a and b) hours after last exposure, the bone marrow samples
prepared according to the method of Sugiyama T. J. natn.
Cancer Inst. 47, 1267–1275, 1971 but with slight
modifications and the slides examined for any chromosome
abnormalities.
Reliability: 2 (valid with restriction)
Comparable to guideline study with acceptable restrictions
no positive control, one sex
Result: Benzene caused a statistically significant clastogenic
effect in all treatment groups.
