Matériels et méthodes III.2.4. Caractérisation des souches bactériennes issues de 12 échantillons de pâtes fermentées prélevées dans 12 ateliers de Ouagadougou III.2.4.1. Préparation des cultures en culot Nous avons repiqué dans chaque tube contenant du bouillon MRS une souche. Nous avons ensuite incubé ces tubes pendant 12 à 24 h (selon la croissance obtenue) à 30°C. Après la croissance nous avons récupéré le culot de bactéries par centrifugation pendant 10 min à 5000 tpm. Ce culot est ensuite lavé et centrifugé deux fois à l’eau distillée stérile. Le lavage permet d’éliminer toutes traces de milieu dans le culot et de ne garder que les souches bactériennes. Enfin, les différentes caractérisations ont été réalisées avec le culot obtenu. III.2.4.2. Coloration Gram La coloration différentielle de Gram a été faite à l’aide du kit prêt à l’emploi de BioMérieux. III.2.4.3. Catalase La caractérisation par réaction enzymatique du type catalase a été faite à partir du kit « ID color catalase » prêt à l’emploi de BioMérieux. III.2.4.4. Galeries API 50 CHL Les profils biochimiques des souches ont été réalisés grâce à l’utilisation de galeries API 50 CHL (BioMérieux) analysés par le logiciel APILAB plus. La classification taxonomique des isolats a été réalisée en établissant une matrice des distances à partir du calcul de l’indice de dissimilitude de Nei et Li. Basé sur cette matrice, un dendrogramme a été établi en utilisant l’algorithme UPGMA (Unweighted Pair Group Method with Arithmetic mean). III.2.4.5.Type métabolique Le type métabolique des souches a été déterminé par dosage des acides acétique et lactique, et éthanol par HPLC (High Performance Liquid Chromatography). III.3. Analyses statistiques Toutes les expériences ont été répliquées trois fois et le dosage des échantillons a été fait en double. L’analyse statistique des données a été réalisée à l’aide du logiciel Statgraphics Plus version 5.1 (Manugistics, Inc., Etats-Unis). Le niveau de signification (P ≤ 0,05) des différences entre les échantillons a été testé par analyse de variance (ANOVA) en utilisant le test des comparaisons multiples de Duncan pour séparer les moyennes (Duncan, 1955). 58
