The early IL-10 resp<strong>on</strong>se in ovine Johne’s disease<br />

The classic early immunological resp<strong>on</strong>se to Mptb is mediated by T-cells with predominant interfer<strong>on</strong> gamma<br />
activity. This resp<strong>on</strong>se is curbed as clinical signs <str<strong>on</strong>g>of</str<strong>on</strong>g> disease appear and may, at least partly, be due to a surge<br />
in <str<strong>on</strong>g>the</str<strong>on</strong>g> activity <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> immunosuppressive cytokine interleukin 10 (IL-10). While <str<strong>on</strong>g>the</str<strong>on</strong>g>re is evidence <str<strong>on</strong>g>of</str<strong>on</strong>g> IL-10 activity<br />
in <str<strong>on</strong>g>the</str<strong>on</strong>g> later stages <str<strong>on</strong>g>of</str<strong>on</strong>g> disease its involvement at earlier time points is not known. The aim <str<strong>on</strong>g>of</str<strong>on</strong>g> this study was<br />
to m<strong>on</strong>itor <str<strong>on</strong>g>the</str<strong>on</strong>g> antigen-specific IL-10 resp<strong>on</strong>se during experimental Johne’s disease. Merino lambs were left<br />
unchallenged (c<strong>on</strong>trols, n=20) or challenged orally with Mptb (n=40). Lymphocytes isolated from blood (every<br />
4 m<strong>on</strong>ths) and lymph nodes (11-13 m<strong>on</strong>ths after challenge) were cultured in <str<strong>on</strong>g>the</str<strong>on</strong>g> presence or absence <str<strong>on</strong>g>of</str<strong>on</strong>g> Mptb<br />
antigen. IL-10 in culture supernatants was determined by ELISA. Faecal shedding and presence <str<strong>on</strong>g>of</str<strong>on</strong>g> Mptb in tissue<br />
samples were determined by culture in a radiometric system (Bactec). Histological lesi<strong>on</strong> type was also assessed.<br />
Antigen-specific IL-10 secreti<strong>on</strong> in peripheral blood <str<strong>on</strong>g>of</str<strong>on</strong>g> sheep exposed to Mptb was significantly higher<br />
than in c<strong>on</strong>trol animals (P < 0.001) as early as 4 m<strong>on</strong>ths p.i. and increased progressively. The peripheral blood<br />
IL-10 resp<strong>on</strong>se in animals with multibacillary lesi<strong>on</strong>s tended to differ from animals with no histological lesi<strong>on</strong>s<br />
or with paucibacillary lesi<strong>on</strong>s. In ileal and jejunal lymph node cells, IL-10 secreti<strong>on</strong> was significantly higher in<br />
<str<strong>on</strong>g>the</str<strong>on</strong>g> exposed animals (P < 0.05). In additi<strong>on</strong>, IL-10 secreti<strong>on</strong> from <str<strong>on</strong>g>the</str<strong>on</strong>g>se cells was significantly higher in animals<br />
with no lesi<strong>on</strong>s or with paucibacillary lesi<strong>on</strong>s when compared to animals with multibacillary lesi<strong>on</strong>s. Our novel<br />
findings dem<strong>on</strong>strate that IL-10 activity occurs so<strong>on</strong> after exposure to Mptb and may play a role in determining<br />
disease outcome.<br />
